Background and objective:1,25-dihydroxy-vitamin D3,(1,25(OH)2D3) is the main active formof vitamin D, the molecular formula C27H44O, molecular weight of384.6377, which can promote intestinal calcium and phosphorus absorptionand deposition.1,25(OH)2D3is used for the treatment of rickets and bonesoftening disease, beside the effect of calcium and phosphorus metabolism,it also can be used for treatment of breast, lung, prostate and other cancers.its anti-tumor effects through the vitamin D receptor-mediated.Because ofgastric tissue distributed the VDR, so1,25(OH)2D3may inhibit humangastric cancer cell proliferation, but the study is less and controversial. Inthis study we dealt SGC-7901cell with1,25(OH)2D3, and observed thegastric cancer cell proliferation and the expression of tumor necrosisfactor α mRNA and protein, to explore the prospects of vitamin D in thetreatment of gastric cancer and possible mechanism, in order to providenew ideas for the chemotherapy of gastric cancer.Methods:1. Human gastric cancer SGC-7901cells were cultured in vitro.2. SGC-7901cells were treated by1,25(OH)-62D3(10,10-7,10-8,10-9mol/L) for24,48,72and96h, respectively.Cell proliferation was measured by MTT assay.3. Measured Cell cycle by flow cytometry (FCM).4. RT-PCR was used to determine the expression of TNF-α mRNA.5. Western blot was used to determine the expression of TNF-αprotein.Results:1,25(OH)2D3significant inhibited SGC-7901cell proliferation(P<0.05),which showed time-dependently and dose-dependently. Treatedwith1,25(OH)2D3for72h,cell cycle showed G0/G1phase arrest(F=9.81,P<0.05). the expression of TNF-α at transcriptional andtranslational level was dose-dependently inhibited t in SGC-7901cells(P<0.05).Conclusion:In vitro,1,25(OH)2D3, significantly inhibit the Human gastric cancerSGC-7901cells proliferation, and can pass in gene transcription of TNFalpha gene level so as to reduce the inhibition of TNF alpha proteinexpression, this tiped1,25(OH)2D3could inhibiting tumor cell proliferationand inhibiting tumor inflammation, improve the cachexia, as well as otherpotential role. So the anti-proliferation effect which was induced by1,25(OH)2D3maybe correlated with down-regulation of TNF-α in gastric carcinoma SGC-7901cell.
