ObjectiveTo establish the optimized technology of separation and purification ofginsenoside Rg1. To investigate the effect of ginsenoside Rg1on theexpression of inflammatory factor in LPS-activated microglia cells.MethodsUltrasonic extraction is used to abstract total saponin ofPanaxginseng. Using silica gel column chromatography to separateginsenoside Rg1. A high performance liquid chromatography method wasestablished for the content of ginsenoside Rg1.LPS-activated microglia cellswere treated for24h in the presence of ginsenoside Rg1. The level of IL-1,NF-kB and iNOS in the culture supernatants were measured byenzyme-linked immunosorbent assay(ELISA).ResultsThe9.91g ginsenoside Rg1was obtained from50g total ginsenoside.The purity was89.63%. The concentration of IL-1, the expression of NF-kBand iNOS in the culture supernatants evidently increased in LPS-activated microglia cells and decreased by the ginsenoside Rg1co-incubated.ConclusionThe technology demonstrate that the method was cheap and the purityis satisfied. So, this technology is suitable to prepare ginsenoside Rg1in lab.Ginsenoside Rg1has neuroprotective functions by inhibiting the expressionof inflammatory factor in activated microglia cells.
