The materials are roots, stem and leaf ginseng from different periods, ginseng roots of15different origins and ginseng roots of4different years. Adopt neutral buffer solution toextract the enzyme solution of Radix Ginseng. Bradford method is applied to determination ofprotein content. Use UV spectrophotometry to test the activities of amylase (AMY),esterase(EST), acid phosphatase (ACP), alkaline phosphatase (ALP), phytase (PHY),Superoxide dismutase (SOD), Glucose Phosphate Isomerase (GPI), Glutamic-pyruvictransaminase (GPT), Fumarase. This reseach in order to provide the theory basis for frtherunderstand the growth status and ginseng growing quality.The results are as follows:(1) The curves of AMY and EST are similar in the process of growth and developmentof ginseng root: Curves show the general upward trend in the period between seedling stageand fruit setting stage, and peak activity appeared in the July5. Between the ripening stageand the withering stage activity is increased and then decreased.then appeared peak activityin the August30. The curves of ACP and ALP are similar in the process of growth anddevelopment of ginseng root: the activity in seedling stage is low, and the peak activityappeared in May15. Curves show the general upward trend after the spreading stage. Activityof PHY was high in the end of spreading stage. The activity of SOD reached the highest in theend of seedling stage. The activity is10-100times to other times. The activity of GPIappeared peak activity in the fruit setting stage. GPT and Fumarase are at a low level ofactivity in seedling stage. The activity of GPT reached peak value in the end of seedling stage.Fumarase appeared peak activity in spreading stage.(2) Activity of AMY, EST, ACP and ALP are similar to the activity dynamics of thestem in the ginseng. They appeared two peak activities between spreading stage and floweringstage, and there is peak value between fruit setting stage and ripening stage. Activity isshowed slow upward trend in root growing stage and after. The activity of PHY is risegradually to peak value in spreading stage, and appeared two peak values in ripening stage.They are1.74±0.39U g-1(FW) and2.26±0.00U g-1(FW). The activity is rising gradually inroot growing stage. The activity of SOD is waved little before ripening stage. The peakactivity appeared in ripening stage and then decreased gradually. The activity of GPI and GPT changes with dynamic similar: The high activity appeared in spreading stage. The peakactivity of the all growing season is appeared in ripening stage,they are214.26±6.13U g-1(FW) and2742.2±41.9U g-1(FW). Then the activity is decreased gradually. Fumaraseappeared two peak activity in spreading stage and flowering stage. The activities in othertimes are waved little.(3) The activity change of AMY, EST, ACP, ALP and PHY ginseng leaves in the process ofgrowth and development are similar. Activity is rising and then decreased between spreadingstage and fruit setting stage. Activity is decline in the ripening stage. Activity is increased atfirst, then decreased in root growing stage. And they are ascended quickly in withering stage.The activity of SOD and GPI changes with dynamic similar: they are rising and thendecreased before fruit setting stage, the activity decreased quickly in ripening stage. Theactivity is waved little in root growing stage. The activity of GPT appeased peak value inspreading stage. It is488.5±52.0U g-1(FW). Activity decreased the lowest in ripening stage。It is24.0±3.9U g-1(FW), and then is risen. The activity decreased little in the end of rootgrowing stage. Fumarase show the general downward trend before withering stage. Theactivity is risen quickly to49.70±0.00U g-1(FW) in withering stage.(4)9kinds of enzyme activity were compared in ginseng from15different areas.Activity of AMY, EST, ACP, ALP, GPT and Fumarase is the highest in ginseng fromWanbao Town of Antu County. Activity of PHY was the highest in ginseng from ErdaogangVillage of Changbai City. Activity of SOD was the highest in ginseng from Beigang Town ofFusong City. Activity of GPI is the highest in ginseng from the Hunchun City. Ginseng from15different areas was clustered into5types.(5) The enzyme activity of4years old ginseng was lower than the5,6and7years oldginseng. Comparison of4,5and6years old ginseng, activity of EST, ALP and GPTincreased with growth period. Activity of GPI decreased with growth period. Activity ofAMY, ACP in7years old ginseng was highest. Fumarase in6years old ginseng was highest.SOD in5years old ginseng was highest.
