Objective:To study the Influence of Electroacupuncture on Gl、GABA in the cerebral cortex and black mass of rats with the spasticity of paralysed limbs after stroke, and study the mechanism of Electroacupuncture’s ameliorating spasm.Methods:Forty SD rats were randomly divided into blank,sham-operation, model,acupuncture technique,Electroacupuncture groups.The blank group were given no manage;the model group were given simulated acupuncture after being model,the acupuncture technique group were needled Yang ling-quan,qu-chu after being model three days successfully,the electroacupuncture group were given electroacupuncture on the basis of the acupuncture technique group.The two groups’ treatmet continue30min,once one day for three days.Observe the changes of rats’neurological damage symptoms and muscle tension.Observe the changes of Glu,GABA and their receptors’ contents in rats’ brain cortex and black mass by Elisa.Observe the changes of mGluRla,GABABRl’ expression in in rats’brain cortex and black mass by RT-PCR.Result:(1)The score of neurological damage symptom:Before treatment,the difference between the blank, sham-operation group and the model group has statistical significance.After treatment,the difference between the two acupuncture groups and the model group has significance.The two acupuncture groups’difference before and after treatment has statistical significance.The difference between conentional acupuncture and electroacupuncture group has no significance(p>0.05).(2)Muscle tension:Before treatment,compared with the blank, sham-operation group,the rats’muscle tension in model,conentional acupuncture, Electro-acupuncture group were increased(p<0.05).After treatment,compared with the model group,the rats’muscle tension in model,conentional acupuncture, Electroacupuncture group were decreased(p<0.05).conentional acupuncture, Electroacupuncture group’muscle tension were decreased compared with pre-treatment(p<0.05).The difference between conentional acupuncture and electroacupuncture group has no significance(p>0.05).(3)G1u、GluR:Compared with the blank, sham-operation group,the content of Glu,GluR in rats brain cortex and black mass were increased in model group(p<0.05).Compared with the model group,the rats’content of Glu,GluR in conentional acupuncture,Electroacupuncture group were decreased (p<0.05). Compared with conentional acupuncture,Electroacupuncture group’content of Glu,GluR were decreased(p<0.05).(4)GABA:①brain cortex:Compared with the blank, sham-operation group,the content of GABA in rats brain cortex were decreased in model group(p<0.05).Compared with the model group,the rats’ content of GABA in conentional acupuncture,Electroacupuncture group were increased(p<0.05). Compared with conentional acupuncture Electroacupuncture group’content of GABA were in creased(p<0.05).②black mass:Compared with the blank, sham-operation group,the content of GABA in rats’black mass were decreased in model group(p<0.05).Compared with the model group,the rats’ content of GABA in conentional acupuncturegroup were increased, but the difference has no statistical significance(p>0.05),the rats’content of GABA in Electroacupuncture group were increased(p<0.05).Compared with conentional acupuncture,Electroacupuncture group’ content of Glu,GluR were increased (p<0.05).(5)GABAR:Compared with the blank, sham-operation group,the content of GABA in rats brain cortex and black mass were decreased in model group(p<0.05).Compared with the model group,the rats’ content of GABA in conentional acupuncture,Electroacupuncture group were increased (p<0.05). Compared with conentional acupuncture,Electroacupuncture group’ content of Glu,GluR were increased(p<0.05).(6)mGluR1amRNA:Compared with the blank, sham-operation group, the expression of mGluR1amRNA in rats’ brain cortex and black mass were enhanced in the model group(p<0.05).Compared with the model group,the expression of mGluR1amRNA were reduced in conventional acupuncture, Electroacupuncture group(p<0.05).Compared with conventional acupuncture,the expression of mGluR1amRNA were reduced in Electroacupuncture group (p<0.05).(7)GABABR1mRNA:Compared with the blank, sham-operation group, the expression of GABABR1mRNA in rats’brain cortex and black mass were reduced in the model group(p<0.05).Compared with the model group,the expression of GABABR1mRNA were enhanced in conventional acupuncture, Electroacupuncture group(p<0.05).Compared with conventional acupuncture,the expression of GABABR1mRNA were enhanced in Electroacupuncture group (p<0.05).Conclusion:Acupuncture can relieve spasm in SCA rats,its mechanism maybe have relationship to regulate Glu,GABA.And Electroacupuncture’s effect is better than conentional acupuncture.
