Establish the model of recurrent spontaneous abortion and a normal pregnancy mice:Mating50female CBA/J mice with20males DBA/2respectively in cage. Inspection to the Yin bolt for0day of pregnancy. Established40recurrent spontaneous abortion and10normal pregnan-cy mice as model respectively.2Grouping and interventionthe model of recurrent spontaneous abortion: Take CBA/J×BALB/c pregnancy mice as a normal pregnancy group (n=10), Take CBA/J×DBA/2pregnancy mice as recurrent spontan-ous abortion group (n=40). According to the order of pregnancy random- ly divided into model group (10only), low dose group of Shou tai pill (10,only3g·kg,d), medium dose groups of Shou tai pill (10only6g,kg,d), and high-dosage groups of Shou tai pill (10,12g only, kg,d).From the first day began to fill pregnancy stomach, give the same amount of saline fill the stomach to a normal pregnancy group and the model group, and the other three were given shoutai pill suspension filling liquid stomach until the14th day. Use immunohistochemical and protein imprinting (Western-blot) method to detection the expression of Annexin A2and TTR.use the SPSS18.0statistical software to processing. Using protein imprinting to detection the expression of Annexin A2Compared with normal group, the expression of Annexin A2in recur-rent spontaneous abortion mice decline significantly,With the interve-ntion of Shou tai pill, expression level of Annexin A2obviously incr- ease, and the most significant group is high dose of Shou tai pill.com-pared with model group, Shou tai pill of high doseã€medium dose and low dose, difference has the obvious significant, they have obvious statistical significance(P<0.01);medium dose and low dose groups compare with h-igh-dosage group, difference has the obvious significant, they have obvi-ous statistical significance(P<0.01).2. Using protein imprinting to detection the expression of TTR Compared with normal group,the expression of TTR Inrecurrent spon-taneous abortion mice decline significantly, With the intervention of Shou tai pill, the most significant increasing group is high dose of Shou tai pill. Effect of Shou tai pill of medium dose and low dose group is not obvi-ous.The modelã€medium dose and low dose group compare with high dos, difference has the obvious significant, they have obvious statistical significance(P<0.01);the medium dose and low dose group compare with model, difference has not the obvious significant, they have not obvious statistical significance(P<0.01).3. Using immunohistochemical to detection the expression of Annexin A2Each group are positive express the tan particles of Annexin A2in the cell membranes,but not in the nucleus.The normal group is more obvious than model group. With the intervention of Shou tai pill,high dose group shading close to normal group,but deeper than medium doseã€low-dose and model group.The model group compared with others,each group were significantly higher than model group.difference has the obvious si-gnificant, they have obvious statistical significance(P<0.01);the medium dose and low dose group Compare with model, difference has the obvious significant, they have obvious statistical significance(P<0.01).4. Using immunohistochemical to detection the expression of Transthyr-etinEach group are positive express the tan particles of TTR in the cell plasma,but not in the nucleus.The normal group is more obvious than model group.With the intervention of Shou tai pill,high dose group shading close to normal group,but deeper than medium doseã€low-dose and model group.compared with the model group, normal group and hi-gh dose were significantly higher.difference has the obvious significant, they have obvious statistical significance(P<0.07);the medium dose and low dose were not significantly higher.difference has not the obvious si-gnificant,they have not obvious statistical significance(P<0.01).compared with the high dose group,the expression of medium and low dose were si- gnificantly lower. difference has the obvious significant(P<0.01),they have obvious statistical significance.Conclusions:The expression of Annexin A2and TTR in recurrent spontaneous abortion mice is lower than a normal group.With the intervention of Shou tai pill, The expression of Annexin A2and TTR in high dose group is increased significantly closing to normal group. The expression of TTR in the me-dium dose and low dose is not obviously increased. difference has not the obvious significant compared with normal group. the expression of Anne-xin A2in the low-dose and the medium dose group also have increased slightly,and have significantly difference compared with model group. They indicate that high dose Shou tai pills can enhance the expression of Annexin A2and TTR in recurrent spontaneous abortion mice. |
