Objective To investigate the effects of inflammatory factors with different concentration to adhesion molecules expression in mesenchymal stem cells derived from human umbilical cord tissue.Methods The MSCs were isolated from human umbilical cord by tissue culture. The expressions of specific markers in MSCs were detected in the physical condition by flow cytometry. The adipogenic and osteogenic induction of MSCs were detected by alizarin and Oil red O staining. MSCs were exposed to TNF-a, IFN-a and IFN-y with different concentration for24h, analyses of expression of CD44, CD49d, CD54, CD58, CD62p, CD62L, CD102and CD106on cell surface were performed using flow cytometry.Results In physiological state, MSCs express extremely low level of CD102, CD106, CD62P, CD62L(<1%) while the expression of CD54was relatively high,41.58%. Stimulated by TNF-a, IFN-a and IFN-y, the expression of CD102, CD106, CD62P and CD62L were increased slightly, but still below10%, while CD54was upregulated with concentration-dependently, up to (70.69±1.08)%. When exposed to TNF-a of40ng/ml, the expression of CD58and CD49d could be upregulated to (52.02±1.35)%,(61.63±0.55)%respectively, CD44could be increased to (61.31±0.71)%when exposed to TNF-a of20ng/ml. CD49d, CD54and CD58were upregulated with IFN-a concentration-dependently, up to (66.36±2.48)%,(76.26±1.85)%and (47.78±0.44)%. When exposed to IFN-a of3×103U/ml, the expression of CD44could be upregulated to(49.8±3.25)%. CD54and CD58were upregulated with IFN-y concentration-dependently, up to (59.66±1.36)%and (43.96±0.62)%, the expression of CD49d could be upregulated to (51.33±0.74)%When exposed to IFN-γ of1×102U/ml, CD44could be upregulated to (73.22±1.93)%When exposed to IFN-y of1×103U/ml.Conclusion TNF-a, IFN-a and IFN-y can elevate signaficatly the expression of CD54, CD49d, CD44and CD58, but not significant for CD102, CD106, CD62P and CD62L on the surface of mesenchymal stem cells derived from human umbilical cord.
