Pick up one strain which was more likely to form Pseudomonas aeruginosa biofilm to do the following research.After treated with different dosages of erythromycin and biyuanshu oral liquid for continuous7days, the Pseudomonas aeruginosa biofilms were observed by AgNO3stainning and the number of viable bacteria were measured by vibrate-serial dilution method.The outcome of the viable bacteria counting is showed by x±SD.Statistical analysis was performed using SAS9.1. Data was analyzed by one-factor analysis of variance using multiple comparison. P was considered to be significant at a level of0.05. Also the already formed Pseudomonas aeruginosa biofilms were treated with different dosages of erythromycin and biyuanshu oral liquid for another24hours, and observed by AgNO3stainning.Result:After cunltured in Tryptic Soy Broth-silicone slides system for continuous seven days,the AgNO3stainning showed that one of the three Pseudomonas aeruginosa strains was more likely to form mature Pseudomonas aeruginosa biofilms,and the result was confirmed by the detetion of SEM.The erythromycin and biyuanshu oral liquid have the effect on inhibit the formation of Pseudomonas aeruginosa biofilms. Under the microscope,the black floccule on the silicone slides was far more less than the positive ones,and the effect was enhanced by the higher concentration.The serial dilution method showed that the viable counts of bacteria of erythromycin and biyuanshu oral liquid treated groups were significantly lower than those non-treated groups(P<0.05).But with the already formed Pseudomonas aeruginosa biofilms the clearance are not significant.Conclusion:1.Using the plate cultue by Tryptic Soy Broth-silicone slides system can establish stable Pseudomonas aeruginosa biofilms in vitro. AgNO3stainning can preliminarily detect the morphology of the Pseudomonas aeruginosa biofilms.
