| Cervical cancer is the second high incidence diseases of women’s malignant tumors inthe word. Currently,85percent of cervical cancer occur in developing countries. Indeveloping countries,13%of female cancer patients are cervical cancer.This incidence isincreasing year by year, and the age of suffering cancer tends to be younger. In developedcountries, due to the effective application of screening and preventive vaccines, theincidence of cervical cancer has declined markedly. But in developing countries, theapplication of effective mearuses are limited,due to poor economy. How to invent a newvaccine,which is inexpensive, efficient and easy to operate, becoming the urgent problem indeveloping countries.A large number of epidemiological data indicated HPV16/18is the main types causeingcervical cancer, accounting for70%of the total. Statistics showed that HPV16mainlycaused cervical squamous cell carcinoma, meanwhile HPV18was the main cause ofcervical adenocarcinoma. A large number of studies found that the malignancy of cervicaladenocarcinoma was higher than that of cervical squamous cell carcinoma, which was earlymetastasis, not sensitive to radiotherapy and chemotherapy, poor prognosis and higherincidence, so early prevention to HPV18infection is particularly important.Attenuated Salmonella as a carrier of DNA vaccine can deliver a variety of antigens, andinduces both cellular and humoral immune response via an easy way by a singleimmunization dose and mucosal route, the safety of which has also been confirmed nearly30years. Therefore, the attenuated Salmonella can be used as a vector to prevent HPV infectioneven achieve the goal of preventing cervical cancer.This study extracted L1gene of human papillomavirus type18from cervix cancertissues of women living in ChangChun area, constructed eukaryotic expression systempVax1-HPV18-L1. Transfected pVax1-HPV18-L1plasmid into BHK cells and detected theexpression product as we expected. Furthermore, we immuned mice with attenuatedSalmonella Ty21a carrying pVax1-HPV18-L1plasmid by intranasal and induced specificantibodies in the sera of mice successfully.For the cervical cancer prevention,this study will provide a new productive idea. |
