Objective:To investigate human cardiac myocytes proliferation and oxidative stress index MOD, SDA and HO-1influence in high glucose environment and in high glucose environment to join adiponectin. Adiponectin may be to the protection mechanism to human myocardial cells, thus offer a new thingking to prevent and treat diabetic cardiomyopathy.Methods:①We divided the human cardiac myocytes we into:normal glucose group,high glucose group and high glucose plus adiponectin group.Cultured in24h,48h,72h respectively②To Measure human cardiac myocytes proliferation activity by MTT test.③By xanthine oxidase to detect SOD activity in the supernatant..④Thiobarbituric acid measured MDA content at different time points in each group in the cell culture supernatant.⑤Different time period each grouping of human cardiac cells heme oxygenase-1(HO-1) expression measured by quantitative PCR technique.Results:1. Compared the OD value in different time and cells by MTT test with high-glucose group and the high sugar plus adiponectin group of24hours, the difference had no statistical significance. After48and72hours, the OD value in high-glucose group was decreased (P<0.05),and compared with high-glucose group, the OD value in high-glucose plus adiponectin group significantly increased(P<0.05).2.The content of SOD decreased in cells cultured with high-glucose compared with normal group,while the content of MDA increased.The indexes mentioned above changes in the opposite direction,respectively (P<0.05).3.The expression of HO-1up-regulated in cells cultured with high-glucose compared with normal group,and it increased more in cells cultured with high-glucose plus adiponectin, respect-ively (P<0.05).Conclusion:The sustanible effects of high-glucose inhabit the cell proliferation,and aggravate the oxidatie stress damage of HCM.Cell proliferation increased and the damage of oxidative stress alleviated with the treatment of adiponectin.Adiponectin protected the cell grow--th in high-glucose.
