Objective1.To clear the specificity of combination with peptide CSNRDARRC and bladder transitional cell carcinoma tissue.2. To explore the subcellular sites peptide CSNRDARRC recognize targetly bladder transitional cell carcinoma tissueMethodsTo choose bladder transitional cell carcinoma (TCC), renal carcinoma, gastric cancer and cystitis glandular paraffin-embedded tissues, there were_respectively107cases,43cases,68cases and16cases.The paraffin-embedded tissues were sliced、 baked、de-paraffinded、retrieved antigen and blocked,then FITC-six amino caproic acid-CSNRDARRC compound was mixed with immunofluorescence technic.The results were analysed qualitatively by fluorescence microscope.The negative control was only mixed with FITC. The datas were analysed by the statistical software SPSS13.0,and there was a significant difference (P<0.05)ResultsAll negative controls did not show highlighted fluorescence points,while bladder transitional cell carcinoma mixed with FITC-six amino caproic acid-CSNRDARRC compound showed highlighted fluorescence points in99cases and the specificity of combination was92.52%(99/107). Renal carcinoma, gastric cancer and cystitis glandular tissues showed highlighted fluorescence points in6cases、5cases、2cases respectively, the specificity of combination was respectively9.52%(5/43),8.82%(6/68),12.50%(2/16).The highlighted fluorescence points were located in nucleusby DAPIstaining. Combination of CSNRDARRC and bladder transitional cell carcinoma has significant difference (P<0.05), compared with renal carcinoma, gastric cancer and cystitis glandular tissues.The combinations of CSNRDARRCand renal carcinoma, gastric cancer and cystitis glandular tissues. have no significant difference.ConclusionCombination of peptide CSNRDARRC and bladder transitional cell carcinoma has higher combination ratio, compared with renal carcinoma, gastric cancer and cystitis glandular tissues,and the binding site of peptide CSNRDARRC is in cell nucleus. It provides theoretical basis for clinic diagnosis of bladder transitional cell carcinoma to peptide CSNRDARRC,and plays basis on therapy of bladder carcinoma.
