Objective To set up rat model of bone cancer pain(BCP) and observe the effect of analgesia by intrathecal injection of HEK293cells stably expressing human preproenkephalin gene in rat model of bone cancer pain and investigate the changes in expression of proteins kinase Cy(PKCy) in the dorsal horn.Methods Forty female adult SD rats(weight180-200g) were randomly divided into4groups(n=10):Naive group (Blank control group in which rats do not do any processing), BCP group(Set up rat model of bone cancer pain in the left tibia in rats. Do not intrathecally inject any drugs.) BCP+HEK293group (Set up rat model of bone cancer pain in the left tibia in rats. Intrathecally injected Ubc-GFP-L.V/HEK293cells.), BCP+HPPE group (Set up rat model of bone cancer pain in the left tibia in rats. Intrathecally injected Plv-Ubc-HPPE/HEK293cells.). Take intrathecal puncture and indwelling catheter in successful models of rats. IN the14th day after operation, BCP+HEK293group and BCP+HPPE group was intrathecally injected Ubc-GFP-L.V/HEK293cell suspension and Plv-Ubc-HPPE/HEK293cell suspension, respectively, once a day, for four days. Paw withdrawal thermal latency (PWTL) were assessed before operation and every3days from the3rd day to the18th day after operation. IN the18th day after operation, after completion of pain assessments, take rat L4-L6spinal cord tissue of each group and detect the expression of PKCy by immunohistochemistry.Results1) Before the bone cancer pain model produced, there was no statistical significant difference of PWTL between the rats in each group (P<0.05).2) From the9th days in the bone cancer model making, Rat PWTL caused by the pain significantly reduced compared with the preoperative(P<0.05). And continue to decrease. At the same time there is statistical difference between Naive group and groups set up rat model of bone cancer pain (P<0.05).3) After the intrathecal injection of cells, the BCP+HEK293group rats PWTL continue to reduce (P<0.05). There is no statistical difference between the BCP+HEK293group and BCP group (P>0.05).
