Objective: This topic application intraperitoneal injection cyclophosphamide (CTX)establish mice bone marrow suppression models, through the observation of the drugon mice before a bone marrow cell count change, using nuclear enzyme-linkedimmunosorbent assay technology (ELISA) detection bone marrow hematopoieticfactor of bone marrow cells on mice granulocyte colony stimulating factor (G-CSF),thrombopoietin (TPO),erythropoietin (EPO) expression level; In mice, the cellculture technology Progenitoring the cell colony granulocyte monocyte progenitor cell(CFU-GM),megakaryocyte hematopoietic progenitor cell (CFU-Meg),erythroidprogenitor cells (CFU-E/BFU-E)and counting, so as to explore the cell colony numberof pure JYK capsule on bone marrow hematopoietic cell proliferation function effecton mice, reliable scientific basis for the mechanism of action of pure JYK capsuleclinical application provides.Methods: Choose clean healthy male Kunming (KM) mice, aged6~8weeks,weight18-22g,according to body weight,the mice were randomly divided into normal group,model group, Tonic Capsule group,Likejun group, JYK capsule high dose group,JYKcapsules medium dose group,JYK capsule low dose group.In the first3days injectcyclophosphamide100mg/kg (normal group injected with normal saline)AM;different drugs l.i.g:0.5%CMC-Na20ml/kg l.i.g to normal group and Modelgroup;Like-Jun32mg/(20ml·kg) l.i.g to LiKe-jun Group;CQFZ Capsule3g/(20ml·kg) l.i.g to CQFZ Capsule Group;JYk Capsule4g/(20ml·kg) l.i.g to JYK highdose group;JYk Capsule3g/(20ml·kg) l.i.g to JYK medium dose group; JYk Capsule2g/(20ml·kg) l.i.g to.JYK low,after3days,only different drugs l.i.g in the afternoon.After modeling8days,mice were sacrificed by cervical,the mice were taken out of thefemur bone marrow cells with normal saline to prepare single cell suspension.Tak-inga part of the single cell suspension,counted bone marrow cells (BMC) with theCytometer.Taken femur serum from the mices,and measured blood growth factorG-CSF,TPO,EPO by enzyme immunoassay kit.Progenitor cells cultured byimplantation in vivo diffusion chamber method and count the cell colony in the lightmicroscope Olympus.Results:①The best result of increasing bone marrow cells is Jing Yuan kang capsulehigh dose group.Jing Yuan kang capsule low dose group increased the role of bonemarrow cells obviously. The results obviously showed that Jing Yuan kang capsulecan promote the recovery of hematopoietic function of bone marrow. Results ofELISA:The effects of JYK capsule on the bone marrow G-CSF,TPO,EPO of miceshowed a dose-dependent.Especially high dose of JYK capsule can improve thehematopoietic function of bone marrow suppression in mice to the normal level.②The results of hematopoietic progenitor cells in vivo culture: Jing Yuan Kang each do- se group could improve bone marrow progenitor cells (CFU-GM, CFU-Meg,CFU-E/BFU-E) colony yields rate of mices. JYK capsule high dose group was moreproduced. JYK capsule can relieve the mices’ bone marrow suppression by CTXinduced. The role of humoral factors may be relevant,Maybe the materials producedby stimulating the body can affect the three progenitor cell proliferation anddifferentiation of certain biologically active substances which play a role.Conclusion: JYK capsule can significantly improve bone marrow suppression causedby CTX,increase bone marrow cells,JYK capsule can adjust feedback incretion incretion of serum G-CSF, TPO, EPOlevels caused by chemotherapy to the normallevel. The results also showed that JYK capsule can also promote the proliferation ofhematopoietic stem cells by humoral factor or some of bioactive materials to increasethree progenitor cells colony. The preliminary experiment confirmed that promotedstem cell proliferation and differentiation may be one of the mechanism of JYKcapsule to improve chemotherapy-induced bone marrow damage and restorehematopoietic function.
