Research On Sperm Concentration On By Adding Astragaloside, Danshen, Progesterone Injection

ObjectiveObjctive To investigate the levels of intraspermic Ca2+under different concentration of Five Seeds Combo, Dodder, Schisandra chinensis, Barbary Wolfberry Fruit, Plantago asiatica, raspberry and progesterone.Specimens collected from115cases of hospital out-patient clinics, were randomly divided into8groups:blank control group, Five Seeds Combo group, Dodder group, Schisandra chinensis group, Barbary Wolfberry Fruit group, Plantago asiatica group, raspberry group, progesterone group, the group joined the contour of the concentration of reagents, Fluo3-AM passive diffusion into the cells, the esterase hydrolysis released Fluo3, Fluo3complexation with calcium ions, the488nm wavelength laser excitation fluorescence arising from its fluorescence and Ca2+was positively correlated by flow cytometry (FCM) with the determination of sperm Ca2+concentration. We undertook statistical analysis using software of SPSS17.0.ResultDetection of progesterone1. FCM, within the RA group sperm Ca2+fluorescence intensity and Salvia, blank group differences were significant (P<0.05), shows that progesterone added in vitro, with astragalus can make sperm Ca2+concentration increased;2. astragaloside group, in the fluorescence intensity increased concentration; RA group significantly lower concentration of fluorescence intensity is weak, with high, medium concentration, there are statistically significant (P<0.05); progesterone group of high, medium and low concentration of fluorescent groups showed no statistical significance (P>0.05).3. LSCM measured astragaloside group, progesterone group compared with the SM group was statistically significant (P<0.05); Danshen group and blank group was no significant difference (P>0.05).ConclusionProgesterone can promote Ca2+influx and intracellular Ca2+concentration increase; high concentration, the concentration of sperm within Five Seeds Combo can promote an increase in Ca2+levels, the same effect with progesterone; Dodder, Schisandra chinensis, Barbary Wolf berry Fruit, Plantago asiatica and raspberry can not be caused by Ca2+concentration changes.
