Objective: In order to investigate the impact of decreased level of B7-H3on cellinvasion and metastasis, we have produced RNAi lentvirus supporter of B7-H3.Additionally, the prostate cell line PC-3was infected by the previous supporter. Finally,we have screened the transformative proportion.Materials and Methods: Using the lentvirus plasmid system, we have screened aPC-3cell lines with a stable and slicnce B7-H3gene. In present study, firstly, usingRT-PCR, we detectd the mRNA expressive level of B7-H3; Additonally, the methods ofmigration and invasion experiment in vitro were utilized to examine and investigate theB7-H3RNA expression and impact of decreased level of B7-H3on cell invasion andmetastasis in prostate cancer.Results: Basesd on the sucessfully infected PC-3cell, the results showed that theexpressive level of B7-H3RNA was declined80%in the screened prostate cancer PC-3cell lines. The migration and invasion experiment revealed that the cells which passed themembrane were less in present cohort compared with the both control cohorts (pvalue<0.05). However, we failed to show a statistical difference between the controlcohorts (p value>0.05).Conclusions: Our findings demonstrated that the declined expressive level of B7-H3in PC-3cell lines inhibit invasion and metastasis, and might serve as a marker in prostatecancer.
