Diagnostic And Prognostic Value Of Serum HE4in Patients With Epithelial Ovarian Cancer

Objectives:In this experimental,serum HE4was determined by enzyme-linked immunosorbent assay(ELISA) in samples from overian tumor patients and healthy female volunteers. Serum CA125level was evaluated at homologous serum specimens for comparison with level of serum HE4,in order to evaluated HE4as a new serum tumor marker for diagnosis of ovarian cancer.Methods:We recruited36primary epithelial ovarian cancer patients,48patients of ovarian benign masses and32healthy volunteers during July2010to July2011from the gynecology-oncology Department of Gansu Provincial People Hospital.3mL peripheral blood was obtained from patients and healthy females by routine venepuncture(vacuum blood collection tube with coagulant).Sample was allowed to clot,centrifuged at3000rpm for15minutes,then,the sypernatant liquid was aspirated and stored at under-20℃until use.Serum HE4and serum CA125concentration was determined by a commercial ELISA.Results:1Serum HE4levels were different between the three groups:ovarian cancerwere significantly higher than the group of benign ovarian tumors and healthy volunteers (P<0.05). However, the HE4was no significant difference between ovarian benign tumor patients and healthy volunteers(P>0.05).2It is no sinificant difference between the histologic subtypes of ovarian cancer(P>0.05).3The serum HE4values measured in stage Ⅲ-ⅣEOC patients were significant higher than in stage Ⅰ-Ⅱ EOC patients(P<0.05).4Serum HE4levels was significant decreased arfer operative than preoperative in patients with EOC(P<0.05).5To assess the clinical potential of HE4and CA125in differentiation EOC from benign ovarian masses:The sensitivity(Se) was proximalis.HE4has higher specificity(Sp),omission diagnose rate(β),positiv predictive value(PV+),correct ratio(π),Youden-index(YI), negative predictive value(PV—),lower mistake diagnose rate(a)and than CA125. Conclusions:1Serum HE4level that was evaluated by ELISA kit of overian cancer were significantHigher than in benign overian tumors and healthy volunteers.2The diagnostic test evaluating showed that serum HE4has a promising sensitivityand specificity. serum HE4has a higher sensitivity and specificity than CA125.These results have identified serum HE4as a potential tumor for overian cancerto increasing the rate of early diagnosis of overian cancer.3The serum HE4after surgery was significantly lower compared with preoperative.That suggest serum HE4may be a useful test to monitor treament response in epithelial Overian cancer.
