Objective:1.The imaging features in two types of oligodendrogliomas were explored for better clinical differential diagnosis and treatment guidance.2. The positive rate of expression VEGF and CD44v6in different levels of Oligodendrogliomas were discussed, in order to understand this tumors from microcosmic.3.Imaging features, expression and Significance of VEGF and CD44v6in oligodendrogliomas, from the perspectivity of molecular biology to understand the types of oligodendrogliomas.Methods:1.59oligodendrogliomas cases in resident were collected, MR and enhanced scan all rows in59cases, samples of59cases after HE, observed in low and high light microscopic and electron microscopic. observation of imaging features and pathological classification of tumor boundary distribution density and tumor cells under the light microscopy,MR signal characteristics and micro-calcification, cystic degeneration,hemorrhage,enhancement intensity and number of vessels under the light microscope.2.59oligodendrogliomas cases were confirmed through pathology, paraffin-embedded tissue samples with formalin (thickness:3um) used standard immunohistochemistry staining, observed the expression of VEGF and CD44v6in different types of OGs.3.60oligodendrogliomas cases in resident from October,2008to January,2012were collected. According to the imaging features of tumors,there are three groups:Group1:elected by the ten cases according to the degree of enhancement; elected by the ten cases according to the boundaries of tumors;elected by the ten cases according to the cystic necrosis of tumors, observe VEGF and CD44v6expression in the number of positive cases separately.4.statistical analysisThe statistical analysis was performed using the SPSS17.0statistical software. According to experimental data, data was examined through X2-test and correlation test, statistical significance was defined as P<0.05. Results:1.comparison of imaging featuresAmong37oligodendrogliomas cases of low grade,the median age is34years old, tumors of29cases were seen in gray-white matter junction of parietal lobe;8cases were frontal lobe;there were22anaplastic oligodendrogliomas (WHO III grade), the median age is32years old, tumors of14cases were seen in gray-white matter junction of parietal lobe,6cases were temporal lobe,2cases were frontal-occipital lobe, focus on observation in the Group:the proportion of cases (WHO II grade) had blurred margin was91.9%(34/37),the proportion of cases (WHO â…¢grade) was4.6%(1/22); the tumor showed hyperintensities on T2-weighted images in37cases (WHO II grade), Among22anaplastic oligodendrogliomas (WHO III grade), the tumor showed mixed intensities on T2-weighted images in3cases(13.6%); significant enhancement in WHO â…¡grade accounted for18.9%(7/37), the WHOâ…¢ grade (77.3%),Among37oligodendrogliomas cases of II grade, peritumoral edema (n=5),cystic degeneration (n=7), hemorrhage (n=8); in WHOâ…¢ grade, peritumoral edema (n=22),cystic degeneration (n=12), hemorrhage (n=9).2.Expression of VEGF and CD44v6in different levels of Oligodendrogliomas In this group, there were37oligodendrogliomas cases of low grade, expression of VEGF:negative (n=25),weak intensity (n=6), moderate intensity (n=3),significant intensity (n=3),positive rate is32.4%; Among22anaplastic oligodendrogliomas (WHO III grade):negative (n=3),weak intensity (n=8), moderate intensity (n=5),significant intensity (n=6), positive rate for86.4%.Expression of CD44v6:in this group, there were37oligodendrogliomas cases of low grade:negative(n=31),weak intensity(n=4), moderate intensity (n=2), significant intensity (n=0), positive rate is16.2%; Among22anaplastic oligodendrogliomas (WHO III grade):negative(n=17), weak intensity(n=3),moderate intensity (n=2), significant intensity (n=0), positive rate for22.7%.3.expression of VEGF, CD44v6in the OGs with different Imaging featuresIn Group1,significant enhancement in10cases, positive expression of VEGF in8cases, the positive rate is80%, positive expression of CD44v6in3cases, the positive rate is30%;mild enhancement in10cases, positive expression of VEGF in3cases, the positive rate is30%,positive expression of CD44v6in1cases, the positive rate is10%.In Group2, blurred margin in10cases, positive expression of VEGF in2cases, the positive rate is20%, positive expression of CD44v6in2cases, the positive rate is20%; clear boundary in10cases, positive expression of VEGF in6cases, the positive rate is60%,positive expression of CD44v6in4cases, the positive rate is40%.In Group3,cystic degeneration in10cases, positive expression of VEGF in5cases, the positive rate is50%, positive expression of CD44v6in1cases, the positive rate is10%; in10cases without cystic degeneration, positive expression of VEGF in4cases, the positive rate is40%, the expression of CD44v6is negative.Conclusion:l.Anaplastic oligodendrogliomas(WHO III grade) had more cystic necrosis,hemorrhage, peritumoral edema and significant enhancement than low grade, which are useful in differentiating the two types of oligodendrogliomas.2.Positive rates of VEGF and CD44v6in OGs are related to the classification, which had a role in preoperative grading, treatment and prognosis.3.Enhanced levels, boundaries and cystic necrosis are involved in expression of VEGF, the tumors with significant enhancement, clear boundary and cystic necrosis had high positive rate of expression of VEGF;the boundary and the invasion of OGs are closely related to CD44v6. |
