| Corydalis Ochotensis Turcz which belongs to the Papaveraceae is always distributed inthe shadow of woods, along the rivers or rocks and deforested regions. In China, it can befound in Liaoning, Jilin, and Heilongjiang province. This herb can be wholly used on sorehurts, dysentery, phthisis, hemoptysis and so on. Generally speaking, there are fewresearches on this herb at home, therefore the research and exploration of the alkaloids incorydalis is very important and practical.1. This article analysed the study on the Papaveraceae and explored its alkaloids whichwill provide a basic preparation for the future research.2. Base on the plenty material and literatures, this paper had an overview of the studieson Corydalis Ochotensis Turcz and the antitumor activity of alkaloids which can be thegroundwork for the future research. The microstructure of its root, stem, leaves and flowerswas drawn. The result of the plant tissue and physicochemical properties showed that theherb contained a lot of chemical composition such as alkaloid, saponin, steroids, saccharidesand phenols. It lays a theoretical foundation for its further development3. This paper has done some research one the extraction and separation of alkaloids inCorydalis Ochotensis Turcz. We use the percolation method with the shattered dry herb. Byusing silica gel column chromatography, recrystallization, TLC and other methods we getfive compounds. Through the13C-NMR,1H-NMR and ESIC-MS result, they are affirmedas Daucosterol(â… ), Ochotensine(â…¡), Protopine(â…¢), Ochotenmine(â…£) and Adlumidine(â…¤).Daucosterol(â… ) is a new discovery in Papaveraceae4. The method of acid dye colorimetry was used to determine the total content of thealkaloid in Corydalis Ochotensis Turcz. The method was decided by the scan of itswavelength and selection of pH of the phosphate buffer and the quantity of acid dye.Precisely measure a certain number of berberine hudrocholride and test solution and thenevaporate it in evaporating dish. Firstly,6ml of citric acid-phosphat bromophenol blue buffer(pH3.0) was used to dissolve the sample and transfer into a separator funnel. Secondly,chloroform10ml was added into the same separator funnel to extract. After one hour’sstanding, the chloroform layer was isolated. anhydrous sodium sulfate0.5g was added todehydrate for30minutes then determine the absorbency of the filterate at415nm. Themethod with bromophenol blue dye was proved to be accurate, stable with good reproducibility in testing experiments. So it was fit for quantitative determination of totalalkaloid in Corydalis Ochotensis Turcz.5. It has been reported that the alkaloids has many pharmacodynamic actions. Someearlier research had already shown that the Corydalis Ochotensis Turcz contains differentkinds of alkaloids. So this paper did a certain antitumor activity in vitro, the result was notvery satisfying, maybe the design of dosage is not suitable. The antitumor of total alkaloidsin Corydalis Ochotensis Turcz needs to be studied further. |
