Objective:①To observe the function of endothelial, oxidative stress and the inflammationreaction in myocardial ischemia-reperfusion injury in rabbits;②To observe the influence ofendothelial function, oxidative stress inflammatory reaction by offering rosuvastatin at varioustime.Method: 30 healthy male domestic rabbits can be divided into 3 groups equally. Group A: theischemia and reperfusion group (group I/R, endured 40 minutes of leaving anterior descendingcoronary artery occlusion, which was followed by 120minutes of reperfusion);Group B:rosuvastatin preconditioning group(group RPre, rosuvastatin (3mg/kg)was injected for 3consecutive days through the posterior auricular vein upon reperfusion, and other steps just asGroup A); Group C: the rosuvastatin postpreconditioning group(group RPost, rosuvastatin (9mg/kg)was injected slowly through the posterior auricular vein upon reperfusion after drawingblood from jugular and other steps just as Group A). After releasing of occlusion, the myocardialischemia-reperfusion model was established by occlusion of left anterior descending coronaryartery for 40minutes evidenced by the elevation of the ST segment≥0.2mv on ECG waveform.ST segment of ECG returned to 1/2 or more of the normal waveform, which indicated thesuccessful reperfusion. The activity of superoxide dismutase and the content of nitricoxide,endothe1in-1, malondialdehyde and interleukin-6 were measured at baseline, which wereat the end of ischemia,60 minutes and 120 minutes after reperfusion.Results:①compared with group I/R, the content of ET-1, MDA and IL-6 of group RPre andgroup RPost were decreased and the content of NO and the activity of SOD were increased at 60minutes and 120 minutes after reperfusion(P﹤0.05);②comparing with group RPost, the contentof NO and the activity of SOD were increased at and the content of ET-1,MDA and IL-6 ofgroup RPre were decreased at the end of ischemia, there was no statistical difference betweengroup RPre and group RPost in 60 minutes and 120 minutes after reperfusion.(P>0.0 5).Conclusion:①with time expansion of myocardial ischemia-reperfusion injury, the function ofendothelial of ischemic coronary works worse and inflammatory reaction becomes more seriousthan ever and oxidative stress turns stronger;②rosuvastatin preconditioning and rosuvastatinpostpreconditioning can improve the function of endothelial of ischemic coronary, reducing thelevel of oxidative stress and inflammation reaction and there are no significant differencebetween the two group in these three aspects;③acute myocardial infarction is much less predictable, so rosuvastatin postconditioning has great clinical significance which is more likelyto be recommended in the reperfusion therapy of acute myocardial infarction in patients.
