Objective:To investigate the effects of clostridium butyricum combined with mesalazine on ulcerative colitis in mice to find its possible mechanism of treating uc.Methods:50specific pathogen free,7-9week-old male BALB/c mice (weight20.0±3.0g), were randomly divided into5groups:normal control group(A), model control group(B), mesalazine group(C), clostridium butyricum group (D), clostridium butyricum combined with mesalazine group (E). A group were fed with distilled water. B group,C group, D group, E group were fed with3%dextran sulfate sodium freely for7consecutive days to establish ulcerative colitis modle. At the same time, group C, group D and group E were orally administrated mesalazine (27.50mg/ml),clostridium butyricum (1×106CFU/ml).mesalazine (27.50mg/ml) and clostridium butyricum (1×106CFU/ml)respectively. During the exprement, general situation such as eating and activities, stool trait, daily weightand occult blood were recorded. On the eighth day, blood was collected from eye vein plexus, separated blood serum by centrifugal machine and stored at-20for ELISA. then the mice of each group were sacrificed, Isolated the whole colon, and taken piece of colon for maldehyde fixed, paraffin embedded, HE staining to observe Colon tissue changes. The following indices were used for evaluation:DAI, colon length and histological injury score. HSP27in colonic mucosa was measured by immuno histochemistry and TNF-ain serum by ELISA method.Results1. BALB/c mice treated with DSS solution developed diarrhea, mucous stool and bloody stool.With days,DAI scores of all Intervention group were increasing. Either clostridium butyricum or mesalazine could reduse the DAI and histological injury scores. Compared with model group, the DAI scores, colon length, histological scores were statistically significant(P<0.05).2. Compared with model control group,in three intervention group, the expression of HSP27in the colonic mucosa was increased (P<0.05) and the expression of TNF-a in the serum was decreased (P<0.05).The effection of mesalazine group was samiliar to clostridium butyricum group (P>0.05).The effection of clostridium butyricum combined with mesalazine group was more effective(P<0.05).Conclution Both clostridium butyricum and mesalazine had therapeutic effects on the acute UC mice clostridium butyricum combined with mesalazine was more effective. Its possible mechanism may be related to up-regulating of HSP27expression and inhibition of TNF-α.
