| ObjectiveTo investigate the changes of cell apoptosis index and The infarct area and the expression of Fas gene and Caspase-3of acute myocardial infarction in rats and the effects of Rosuvastatin on them.To investigate the effect of Rosuvastatin on the cell apoptosis of acute myocardial and the mechanism.Materials and MethodsRat models with acute myocardial infarction were performed by ligating left coronary artery(LCA) in40male SD rates.24hours later,32survival rates were randomly divided into two groups:group statin and group control.Group statin (n=16) was gavaged with Rosuvastatin lmg/kg/day, group sham(n=9) and group control (n=16)were gavaged with equal volume of saline from the first day after myocardial infarction. One month later, concentration of the infarct area, the apoptosis index, expression of apoptosis gene Fas and caspase-3were compared with each group(group sham=9,group statin=12,group control=10). the infarctarea were defined with Evans blue and TTC staining.Transferase-mediated dUTP nick end labeling(TUNEL)and Immuno-histochemical staining(SP) were used respectively to detect the changes of apoptosis and protein expression of Fas and caspase-3; mRNA expression of Fas gene and Caspase-3were evaluated by RT-PCR analysis.ResultsThe apoptosis index, the infarct area, positive indexs of Fas and Caspase-3proteins, mRNA expression of Fas and Caspase-3genes were all increased in group statin and group control compared with group sham(P<0.05the difference is significantly).All that were decreased in group statin compared with the group control(.P<0.05the difference is significantly).ConclusionsCell apoptosis index, the infarct area, positive indexs of Fas and Caspase-3proteins, mRNA expression of Fas and Caspase-3genes are increased in acute myocardial infarction in rats.Rosuvastatin reduce the myocardial cells apoptosis of acute myocardial infarction rats, The main mechanism is by reducing the expression of Fas and Caspase-3, which attributed to the cell apoptosis index and infarcted area decrease, improve Cardiac remodeling and cardiac function. |
