OBJECTIVE To prepare the aqueous extracts and alcoholic extracts of pangolin. To compare the analgesic effect between aqueous extracts and alcoholic extracts of pangolin. To study the analgesic effect, the anti-inflammatory effect, the acute toxicity reaction of pangolin alcoholic extracts.METHODS (1)Drug preparation:The aqueous extracts and alcoholic extracts of pangolin were prepared by two different methods.(2)The comparative study of analgesic effect between aqueous extracts and alcoholic extracts of pangolin:After intragastric administration with the dose of3.00g/kg,1.50g/kg,0.75g/kg for mice, the analgesic effect comparison between aqueous extracts and alcoholic extracts of pangolin was studied by Hot-plate test and acetic acid-induced writhing test in mice.(3)The experimental study of analgesic effect:After intragastric administration with the dose of3.00g/kg,1.50g/kg,0.75g/kg for mice and the dose of2.09g/kg,1.04g/kg,0.52g/kg for rats, the analgesic effect and mechanism of the alcoholic extracts was observed by the Hot-plate test in mice, the tail-flick test in mice, the acetic acid-induced writhing test in mice, the test of carrageenan-induced inflammatory model in rats, and the content of NA, PGE2in serum, PGE2in the phlogogenic legs of the carrageenan-induced inflammatory model of rats.(4)The experimental study of anti-inflammatory effect:After intragastric administration with the dose of3.00g/kg,1.50g/kg,0.75g/kg for mice and the dose of2.09g/kg,1.04g/kg,0.52g/kg for rats, the anti-inflammatory effect and mechanism of the alcoholic extracts was observed by the test of carrageenan-induced voix pedis swelling in rats, the test of xylene-induced auricle swelling in mice, and the content of SOD, MDA, GSH-Px in serum of the carrageenan-induced inflammatory model of rats.(5)The acute toxicity test of pangolin alcoholic extracts:The concentration of pangolin alcoholic extracts was0.30g/ml. In this test, the toxic symptoms and mortality were recorded, and the maximum tolerate dose of the alcoholic extracts was studied after intragastric administration with the dose of0.4ml/10g.RESULTS (1)Drugs preparation:With the extraction rate of7.54%and10.25%, respectively, the dry powder of aqueous extracts and alcoholic extracts of pangolin were prepared successfully.(2)The comparative study of analgesic effect between aqueous extracts and alcoholic extracts of pangolin:Both aqueous extracts and alcoholic extracts of pangolin could prolong the pain threshold of mice in the Hot-plate test(P<0.05or P<0.01), and could decrease the writhing time in the acetic acid induced writhing test(P<0.05or P<0.01).(3)The experimental study of analgesic effect:The pain threshold was prolonged by alcoholic extracts in the Hot-plate test, the tail-flick test, and the model of carrageenan-induced inflammatory of rats (P<0.05or P<0.01); the writhing times was decreased by alcoholic extracts in the acetic acid-induced writhing test of mice(P<0.05or P<0.01); the content of NA, PGE2in serum, PGE2in the phlogogenic legs was decreased by alcoholic extracts in the model of carrageenan-induced inflammatory of rats(P<0.05or P<0.01), the analgesic effect and the dose had dose-effect relationship.(4)The experimental study of anti-inflammatory effect:The alcoholic extracts had the inhibiting effect both on the carrageenan-induced voix pedis swelling in rats and xylene-induced auricle swelling in mice(P<0.05or P<0.01); in the model of carrageenan-induced inflammatory of rats, the content of SOD and GSH-Px in serum was increased by the alcoholic extracts, while the content of MDA in serum was decreased(P<0.05or P<0.01), the strength of anti-inflammatory effect and the dose had dose-effect relationship.(5)The acute toxicity test:With24g/kg intragastric administration everyday, the mice lessened in activity, among20mice,6mice showed tachypnea, but got better after15minutes, no one died or showed adnornal during the experiment(7days),so failed to calculate the median lethal dose (LD50).The experiment showed that the maximum tolerate dose of the alcoholic extracts in mice was higher than234.24g/kg, which was1639.68times as much as the administration for man everyday in clinic.CONCLUSIONS (1) The dry powder of aqueous extracts and alcoholic extracts of pangolin are prepared successfully, and the extraction rate of alcoholic extracts is higher.(2) Both aqueous extracts and alcoholic extracts show the analgesic effect, but the analgesic effect of alcoholic extracts is stronger, the effect begin more rapidly, the duration is much longer.(3)The alcoholic extracts of pangolin have the effect of analgesic, the effect and the dose have dose-effect relationship, and the mechanism is concerned with the decrease of NA and PGE2.(4)The alcoholic extracts of pangolin have the effect of anti-inflammatory, the effect and the dose have dose-effect relationship, and the mechanism is concerned with the increase of SOD,GSH-Px, and the decrease of MDA.(5)The acute toxicity test show that alcoholic extracts of pangolin is low toxicity, wide safety range.
