| Objective:To research the dynamic change of interleukin-17(IL-17) in peripheral blood at different time intervals after the mouse were infected by Blastocystis hominis, as well as the expression of IL-17and interleukin-23(IL-23) in BABL/C mice’s intestinal mucosa. To explore the relationship between IL-17and IL-23, as well as the function and mechanism of immune response of IL-17in infection with Blastocystis hominis. To provide a new basis and thread with development of disease, pathogenesis and treatment in infection with Blastocystis hominis.Methods:Blastocystis hominis were cultivated in vitro by IMDM culture medium,120BABL/C mice were randomly divided into different groups:experimental group, group with immunosuppressant and normal group. For the experimental group, the method of intragastric infusion was adopted to infect BABL/C Mice1000-seed weight×107per mouse; for the normal group and group with immunosuppressant, BABL/C mice were fed with equal physiological saline and was fed by SPF.4BABL/C mice from each groups were randomly sacrificed on the day2,3,4,5,6,7,8,10,12and14post infection, respectively. ELISA was used to measure the expression level of IL-17in peripheral blood at different time intervals of these mice. Duodenum, jejunum, ileum and colon were simultaneously taken out in preparation for tissue section which from3group’s mice on the day5post infection. The expression of interleukin-17(IL-17) and interleukin-23(IL-23) in different part of bowel mucosa were determined by immunohistoehemistry analys.Results:The level of serum IL-17of immunosuppressant group and normal group maintained relatively be low through out the Emzyme linked immunosorbent assay (P>0.05), the expression of serum IL-17in the two group’s mice were no statistical significance in the same time (P>0.05); The level of serum IL-17of experimental group began to rise after infection, and IL-17concentration in the serum of mice raised to the highest point at day5, then decreased. Compared with the normal group and immunosuppressant group, the level of serum IL-17in experimental group showed a great increase at day4, day5and day6, and there was a statistical significance (P<0.05), The expression of the other times was no significant (P>0.05).The Immunohistochemical analysis results in mice’s intestinal tissue showed that the positive cells of IL-17and IL-23were distributed mainly intestinal mucosa lamina propria, in the epithelial cells and lamina muscularis also have some positive cells express occasionally, and the positive staining was mainly located in cytoplasm. The expression of IL-17and IL-23in mice’s intestinal mucosa (duodenum, jejunum, ileum and colon) in experimental group, whici was significantly higher than the group with immunosuppressant and normal group (P<0.05). The expression of IL-17in mice’s duodenum, jejunum and colon were significantly higher than ileum in experimental group (P<0.05). The expression of IL-23in mice’s duodenum and jejunum and colon were significantly higher than ileum and colon in experimental group (P<0.05).Conclusion:1. IL-17were highly expressed in the peripheral blood and intestinal mucosa of mice infected with Blastocvstis hominis. IL-17involved in the immunomodulatory effects with Blastocystis hominis infection, which played an important role in the prophase of parasitic infection, but also cause the organism to produce certain pathological damage;2. IL-23was highly expressed in the intestinal mucosa of mice infected with Blastocvstis hominis, there was similarities with the expression of IL-17. IL-23may also be involved in the immunomodulatory effects with Blastocystis hominis infection, which played an mutual regulatory role with IL-17. |
