Objective:To preliminarily observe the pathogenesis of the diabetes cardiomyopathy through observing the myocardial changes of Caveolin-3(Cln-3) in type1diabetic rats,type2diabetic rats and high-fat diet rats.Materials and Methods:Fifty male SD rats were randomly divided into nomal control(NC) group, high-fat diet group,Type1Diabetes(T1DM) group and Type2Diabetes(T2DM) group. Rats in NC group and T1DM group were fed with normal diet, the high-fat diet group and T2DM group were fed with high-fat diet. Three weeks later,T1DM group were provided STZ(65mg/kg) by intraperitoneal injection to induce T1DM, T2DM group underwent injection with streptozotocin (25mg/kg). NC group and high-fat diet rats were injected with sodium citrate.Citrate buffer solution,all groups were continued fed with appropriate diet. After4weeks, the biochemical changes of serum and myocardial tissue were measured, the total protein content of Cln-3was analyzed by Western blot.Results:(1)Compared with the NC group,LDH(P<0.05),CK(P<0.05),MDA(P<0.05), FINS(P<0.01) contents in high-fat diet group,T1DM group,T2DM group and FBG (p<0.01),HOMA-IR(P<0.01) in T1DM group,T2DM group were signify-cantly increased;SOD(p<0.01) and ISI (p<0.01) were significantly decreased in T1DM group and T2DM group.Compared with high-fat diet group, FBG (p<0.01),CK(p<0.01),HOMA-IR(P<0.05) were significantly increased,ISI (p<0.01) were significantly decreased and LDH,MDA,SOD,FINS had no signi-fycant difference in T1DM group,T2DM group; compared between T1DM group and T2DM group,all the parameters had no significant difference.(2) Western blot showed that:Compared with NC group, the expression levels of Cln-3had no significant difference in high-fed diet group,were significantly increased(p<0.05)in T1DM,T2DM group. Compared with in high-fed diet group, the expression levels of Cln-3were significantly increased (p<0.05),compared between T1DM group and T2DM group,the expression levels of Cln-3were significantly increased (p<0.01)in T1DM group.Conclusions:(1) Diabetic myocardial injury is happened in rat models of T1DM and T2DM in this experiment.(2) Myocardial injury may be due to the increased generation of free radical, increased levels of lipid peroxidation and insulin resistance in T2DM rats.(3) The expression levels of Cln-3is significantly increased in injury myocardium in T1DM and T2DM models.
