Objective To evaluate the osteogenetic capacity in vitro of HUCMSC transfected the tet-on lentiviral vectors carrying hBMP-2.Methods HUCMSCs which was third generation were cultured and divided into4experiment groups:stably transfected HUCMSCs added doxcycline; stably transfected HUCMSCs without doxcycline; HUCMSCs transfected by GFP; untransfected HUCMSCs. RT-PCR, alizarin red staining, and ALP activity assay and staining were performed.Results In the stably transfected HUCMSCs group, we could notably detect the expression of hBMP2, and the alizarin red and ALP staining were greatly positive. In the stably transfected HUCMSCs without doxcycline, we could detect little expression of hBMP2, and the alizarin red and ALP staining were weakly positive. In the remained two groups there were no positive results.Conclusion Tet-on-regulated lentiviral vector containing hBMP-2could infect HUMSCs with high efficiency successfully, and the inducible expression of hBMP-2and the osteogenetic capacity were researched, which provide an effective and simple method for the further study of stem cells and animal experiment in vivo. |
