Fructus Corni fruit core is the dried core of the ripe fruit of Cornus OfficinalisSieb.Et Zucc, which is three times heavier than the pulp. The pulp of Fructus Cornifruit is a traditionary and valuable Chinese medicine, and is commonly used to treatsome clinical disease. But the core of Fructus Corni fruit is discarded as garbage,which makes a big lose. It is discovered that the core and the pulp are resemble eachother in component, while the content is different. Besides, some nutritional ingredientin Fructus Corni fruit core is better than in the pulp. All these show that Fructus Cornifruit core is valuable and useful. So far, research on chemical composition andpharmacological effect of Fructus Corni fruit core is little.Objectives: To study some problem about the antioxidative activity of fructuscorni fruit core.Methods:1. Fructus Corni fruit core extraction was prepared by resin enrichment andsolvent extraction two methods; Determine the content of total polyphenol in FructusCorni fruit core extraction with Folin-Ciocalten method, using propyl gallate as thestandard substance.2. Study the differences of chemical constituents between the core and pulp ofFructus Corni, extractions by different methods with HPLC-PDA method.3. Anti-oxidative activity in vitro was determined by adopting total reducingpower, the effects of sodium nitrite scavenging and DPPH free radical-scavenging asthe indexes.4. Anti-oxidative activity in vivo was determined by assaying the activity of totalsuperoxide dismutase(SOD), catalase(CAT), glutathione peroxidase(GSH-PX) and thecontent of malonaldehyde(MDA) in the plasma and liver tissue. 5. Fructus Corni fruit core extraction was separated and purified by usingdifferent kinds of medium such as Polyamide, Diaion HP20and SephadexTMLH-20.The structures of compounds were elucidated by NMR and Thin LayerChromatography.Results:1. Six extractions were obtained by two methods; The content of total polyphenolin extraction was high, the content of total polyphenol in10%and30%methanolextraction was as high as about70%.2. Constituents in Fructus Corni fruit core were far more than in Fructus Cornifruit pulp, and the compounds with galloyl were importent constituents in FructusCorni fruit core; Constituents in Fructus Corni fruit core extractions prepared bysolvent extraction were different, but parent nucleus in these extractions was same.3. Total reducing power of six extractions was strong; The extracts can alsoscavenge sodium nitrite.30%methanol fraction and Butanol extraction presented ascavenging rate for sodium nitrite above80%when the concentration was0.6mg·mL-1;Six extractions exhibited higher DPPH free radical-scavenging ability than Vc.4. The extraction of the Fructus Corni fruit core can increase the activity of SOD,CAT, GSH-PX and decrease the content of MDA by orally administered to aging mice.5. Four compounds were obtained from Fructus Corni fruit core extraction, theywere betulinic acid, gallic acid,6-O-galloyl-β-D-glucopyranose and1-O-galloyl-β-D-glucoside.Conclusions: There are a lot of polyphenols compounds in Fructus Corni fruitcore, Constituents in Fructus Corni fruit core were far more than in Fructus Corni fruitpulp, and the compounds with galloyl were importent constituents in Fructus Cornifruit core; Anti-oxidative activity in both vitro and vivo is better. The actual value ofFructus Corni fruit core is high. |
