The Effect Of Atorvastatin On Myocardial Fibrosis In Rabitts Model Of Diabetes Mellitus

1ObjectiveTo study the content of collagen fibre in myocardial tissue and assess diastolicand systo1ic ventricular function of the rabbits with type2diabetes, evaluate theeffect of atorvastatin in myocardial fibrosis in rabbit model with diabetes mellitus.2Methods22male New Zealand white rabitts were randomly divided into threegroups:normal group(n=6),DMgroup(n=8), atorvastatin group (n=8), atorvastatin5mg/kg/d,TG, TC, GLU,BUN,Cr contents in the serum were mearsured in4week,16week. Collage content was observed by VG staining. The cardiac functionwas examined by echocardiography and TDI in each group respectively.3ResultsCompared with normal group and atorvastatin group, GLU content in DM groupwas significantly increased (P<0.01). Compared with the normal group, The amountof collagen fibre was increased in diabetic rabbits myocardium(P<0.05). Comparedwith the normal group, A ratio increased while E/A ratio decreased in DM group(P<0.01). Compared with the DM group, The amount of albuminuria wassignificantly decreased in atorvastatin group (P<0.01).4ConclusionALX（50mg／kg+100mg／kg）intraperitoneal injection after sustained highglucose and high fat diet4weeks is the optimal method for inducing rabbit diabeticmellitus. Characteristics such as stabilization of blood glucose, high success rate ofmodel construction, low mortaliy，and a series of cardic and renal complications, canbe seen in DM group rabbits. Diastolic function has been impaired before thedevelopment of myocardial systolic dysfunction in early-stage of diabetesmellitus.Independently of its cholesterol-lowering apacity,atorvastatin reduced thedevelopment of myocardial fibrosis.
