Cervical cancer is one of the most common female cancers, in recent years herb isused more and more widely in treating the cervical cancer for its superior anti-cancereffect. Scutellaria baicalensis Georgi is the common traditional Chinese medicine, and itsactive ingredients can inhibit the growth of many kinds of cancer cells. However, theanti-cervical cancer effect and mechanism of Scutellaria baicalensis Georgi has not beenclarified in detail.In order to study the effect and mechanism of Scutellaria baicalensis Georgi oncervical cancer and explore the active anti-cancer ingredients of Scutellaria baicalensisGeorgi, the ethanol extract 1 of Scutellaria baicalensis Georgi and ethanol extract 2 ofScutellaria baicalensis Georgi was extracted and separated from the Scutellariabaicalensis Georgi powder, and being used on the U14 cervical cancer mice model. Thetumor inhibition rate, tumor cell morphology, tumor cell cycle and apoptosis, and theexpression of Bcl-2 and Bax were detected in this research. In addition, the thymus andspleen index, the content of TNF-αand IL-8 in the serum of tumor-bearing mice weredetected to study the immune system effect of ethanol extract of Scutellaria baicalensisGeorgi on U14 cervical cancer tumor-bearing mice. The results showed that ethanolextract 1 of Scutellaria baicalensis Georgi and ethanol extract 2 of Scutellariabaicalensis Georgi could inhibit the tumor growth of U14 cervical cancer tumor-bearingmice, and the possible mechanism was causing tumor cell cycle arrest at S phase,decreasing the expression of anti-apoptotic gene Bcl-2, increasing the expression ofpro-apoptotic gene Bax, and inducing the tumor cell apoptosis. In addition, ethanolextract 1 of Scutellaria baicalensis Georgi and ethanol extract 2 of Scutellariabaicalensis Georgi could also increase the immune organs index and the TNF-αcontentof tumor-bearing mice, and reduce the IL-8 content, so that to enhance the immunefunction of tumor-bearing mice and improve the anti-tumor ability of mice.This research was based on the U14 cervical cancer mice model, and explored theanti-cervical cancer effect and mechanism of Scutellaria baicalensis Georgi from apoptosis and immune aspects. The study initially reveals the mechanism of ethanolextract of Scutellaria baicalensis Georgi on inducing apoptosis of cervical cancer cellsand enhancing the immune function, and laies the foundation for exploring the activeanti-cancer compounds of Scutellaria baicalensis Georgi, it also provides a scientificbasis for the development of clinical anti-cervical cancer herb. |
