According to cloning standards and screening SRBR Green PCR system, quantitative real-time RT-PCR method suitable for studying expression revels of StAR、CYP11A1、CYP17A1、CYP2E1、CYP2A19、CYP2C34mRNA was established and used to evaluate the feasibility of experimental miniat ure pigs’livers and testis tissue using as model of security estimate of drug by quantitation e xpression livers of boar taint gene in livers and testis tissue of8-9months old Bama miniature pigs, Tai Hu pigs, Landrace pigs, Yorker pigs and Rongchang pigs. Then further study of smell genes in the same breeds (Bama) at different ages (newborn, January, February, March, April, June, one year) in the liver and testis tissue expression. And determination of hormones in the blood; Finally, phenotypic analysis of boar taint substances androstenone and skatole content concentration in muscle and plasma. The following was concluded after comparing these results to that of human counterpart documented:1. The genes of StAR、CYP11A1、CYP17A1、CYP2E1、CYP2A19、CYP2C34play an important role in the development of boar taint. The gene of CYP2E1、 CYP2A19、CYP2C34were highly expressed in the liver of Landrace, Yorker and BamaMinipig, whose age were between8and9months old. Gene expression of CYP11A1、CYP17A1、StAR were highly expressed in the testis of rongchang pig and Taihu pig. The level of Testosterone and Estradiol in the blood was different in different breeds.and the two highest are rongchang pig and Taihu pig.2. BamaMinipig hepatic expression of CYP2E1、CYP2A19、CYP2C34were highest at the age of3months.the gene of CYP11A1、CYP17A1、StAR expressed highest by one year old. It took3、12months to maximize the secretion of Testosterone and Estradiol.3. In different pig breeds, through phenotypic analysis of boar taint substances androstenone and skatole content in muscle and plasma concentration, we found that the Rongchang pigs and Taihu pigs muscle and blood androstenone (2.9028±0.1342 ng/mL,3.2097±1.1023ng/mL) and ((2.6814±0.2484ng/mL,2.8718±1.2361ng/mL) and skatole prime (29.7182±1.1461ng/mL,37.4997±1.8394ng/mL) and (39.8411±2.3543ng/mL,21.0334±1.6803ng/mL) were higher than Landrace pigs,York pigs and Bama miniture pigs. The level of Androstenone in the blood and muscle were both high in rongchang pig and Taihu pig,the results of skatole were similar. Androstenone and skatole levels in the blood and muscle increased with age in BamaMinipig. To age of June muscle content to3.3889±0.7451ng/mL, plasma the androstenone content1.1789±0.1234ng/mL. At the same time, androstenone regression rate of0.9478, the the skatole regression rate of0.9999.In summary, conclusions were as follows:Lhepatic expression of CYP2E1、CYP2A19. CYP2C34can affect the level of skatole,And the level of Androstenone in testis tissue can be regulated by the gene of CYP11A1、CYP17A1、StAR.2.androstenone and skatole levels increased with age in Bama entire male pigs, and were dose related with boar taint1, there were different boar taint in different breeds of pig.
