Temperature is the key ecological adaptability factor in the growth and development of the blueberries.Cold hardiness and chilling requirement are main factors that limit the blueberry area expanded. This study use our unique wild Vacinium resource, develop cold hardiness and chilling requirement traits associated SNP markers with the existing blueberry cultivars and interspecific Fi hybrid, molecular marker-assisted breeding. The main findings are as follows:1.Blueberries cold hardiness and chilling requirement phenotypic identificationDetect Northland, Chippewa, Duke, Bluecroup, Darrow, Powderblue6varieties1-year-old branch conductivity, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were measured to determine the cold resistance,using≤7.2℃models germination rate statistics determine chilling requirement level.2.Blueberries artificial hybridization pollination test and hybrids breeding preservationSelect Darrow, Bluecrop, Duke, Chippwa, Northland, Powderblue6varieties cross test,get1183hybrid fruits. Summed up the blueberries artificial pollination hybridization, retained more than20,000hybrid seeds. Take seedling nursery trials, summed reproductive technologies.3.The development, screening, and validation of the SNP markers associated with blueberries cold hardiness and chilling requirement traitsSelect Bluecroup, Powderblue, Sharpblue, Blomidon and two of our unique wild Vacinium resource for the development of SNP markers. Molecular markers with NCBI EST sequence and Blueberries EST database comparison. Detect authentication in75blueberries groups DNA, determine the cold hardiness traits SNP marker SL8088, the chilling requirement traits SNP marker SL16592.4. Molecular marker-assisted breedingUse SNP markers associated with cold hardiness and chilling requirement traits for early identification of29DNA in F1generation seedlings.
