Xichuan Silky Fowl originated in Xichuan County, Nanyang City, Henan Province, has thecharacteristics of the "bird beak, black legs, black skin, black bone, black meat" and has high ratioof green shell eggs (73%). Now it has declared national local chicken germplasm resources andobtained the local chicken germplasm resources authentication in henan province. In this study,variation of eggshell Pigment in different ages of earlier laying period of Xichuan Silky Fowl hasbeen researched, and its eggshell pigment category has been verified. Genetic Diversity of thisgroup has been studied by Microsatellite Markers, and compared with Silky Chicken andDongxiang Blue-shell Chicken. The Association of Microsatellite Markers with22ApparentCharacters has also been analyzed. SNP analysis for heme oxygenase-1of Blue-egg related geneand Association analysis with Egg Quality Traits has been done to verify the genetic pathways andmetabolic mechanism.The foiiows was the results:(1) Take the eggshell pigment contentdetermination of green shell eggs in the four weeks of23weeks,25weeks,27weeks,31week which were produced by the xichuan silky,the resultashowed that: The absorption curve of eggshell solution between Xichuan Silky chicken and otherstudied breeds were similar.There are obvious peak between410nm-420nm,beside,the highestpeak was appear between410nm-580nm.also,there are another small peak at680nm, which wasconform to description in the reports of protoporphyrin and biliverdin’s spectra.so,the researchexplain that the main components of the xichuan silky eggs shell pigment is protoporphyrin andbiliverdin.(2)Compare the UV spectrophotometric absorbance value of eggshell pigments in thedifferent age,we found that,the27weeks,31weeks was significantly higher than the eggshellspigment content of23weeks and25weeks(P<0.05).We also found that,the biliverdin by theincrease of the age.the content of protoporphyrin was increased after31weeks declined,but still slightly higher than the first two batches(23weeks and25weeks).(3)Take the Analysis of Population Genetic Diversity between the Xichuan Silky, Silky fowland lushi green eggshell chicken in16microsatellite markers,we found that:there are difference inthe same locus and genotype between three colonies. The average effective number of alleles withthe Xichuan Silky was3.2,the Silky fowl was3.4,the lushi green eggshell chicken was3.3. thegroups heterozygosity of Xichuan Silky was0.2089-0.8305, the groups heterozygosity of Silkyfowl was0.4884-0.8162, the groups heterozygosity of lushi green eggshell chicken was0.4765-0.7953;the average heterozygosity(HK) of Xichuan Silky was0.661, the averageheterozygosity(HK) of Silky fowl was0.690, the average heterozygosity(HK) of lushi greeneggshell chicken was0.675; the polymorphic information content of Xichuan Silky was0.1198-0.7995, the polymorphic information content of Silky fowl was0.1852-0.7608, thepolymorphic information content of lushi green eggshell chicken was0.2035-0.7641; the average polymorphic information content(PIC) of Xichuan Silky was0.515, the average polymorphicinformation content (PIC)of Silky fowl was0.523, the average polymorphic informationcontent(PIC) of lushi green eggshell chicken was0.537. In addition, the most of16microsatellitemarkers in Xichuan Silky were moderate or high polymorphism,the PIC was rich,thischaracteristics was in accordance with natural population which were closed.(4)Analyzed the genetic distance and genetic similarity between three groups with16microsatellite markers, wherein,the genetic distance between silky chicken and Lushi greeneggshell chicken was0.1154,the genetic similarity was0.8910;the genetic distance betweenXichuan black-boned chicken and silky chicken was0.2210,the genetic similarity was0.8017; thegenetic distance between Xichuan black-boned chicken and Lushi green eggshell chicken was0.2116, the genetic similarity was0.8093; by contrast,the genetic distance between Xichuanblack-boned chicken and Lushi green eggshell chicken was more recent.(5)Selected14gene sites which have higher polymorphism information content from16microsatellite markers, and make association analysis with apparent traits. The results show that:Genotype of Site LEI0194was significant correlation(P<0.05) with eggshell strength、Chestdepth; Genotype of Site LEI0192was highly significant correlation(P<0.01)with yolk weight;Genotype of Site MCW206was highly significant correlation(P<0.01)with yolk color; Genotypeof Site ADL0137was highly significant correlation(P<0.01)with egg ransverse diameter;Genotype of Site LEI0073was significant correlation(P<0.05) with egg ransverse diameter、eggweight、chest width; Genotype of Site ADL0136was significant correlation(P<0.05) with eggransverse diameter、shank girth、toe length; Genotype of Site ADL0143was significant correlation(P<0.05) with middle part eggshell thickness; Genotype of Site ADL185was significantcorrelation(P<0.05) with protoporphyrin content、biliverdin content、pelvis breadth; Genotypeof Site ADL260was significant correlation(P<0.05) with yolk weight; Genotype of SiteMCW0276was highly significant correlation(P<0.01)with biliverdin content、keel length、pelvisbreadth.(6) We selected15clear-genotypestatus in16microsatellite markers, Analyzed with theeggshell color traits of Xichuan black-boned chicken group,the result said,MCW0206, LEI0073,ADL260, MCW0276, ADL225five sites was significant correlation with eggshell color exists(P<0.05), in which ADL260locus was a highly significant correlation (P<0.01).(7) SNP search for heme oxygenase‐1gene of Xichuan Silky Fowl group was analyzed, andtwo SNP sites were searched in this study, association analysis with green-eggshell characteristicsindicated that there were all no significant relativity.(8) Association of gene frequency of the two SNP sites with Egg Quality Traits was analyzed,there was no significant relativity(P>0.05)for SNP1site, there was a significant correlation(P<0.05)for SNP2site with yolk weight.
