The rice blast caused by filamentous fungus Magnaporthe oryzae is one of the most important diseases affecting global rice production. Furthermore, the study of disease-caused basis of M.oryzae is not only beneficial for rice blast control, but also served as a representative model system of revealing other fungal pathogenesis and pathogen-plant interactions.In eukaryotic cells, cell activities are regulated by a variety of signaling pathways and signaling transduction is often carried out through phosphorylation and dephosphorylation. Phosphorylation of phosphatidylinositol plays an important physiological role in regulating cell growth, differentiation, cytoskeleton reconstruction, Ca2+transference and membrane transportation. RAS-cAMP signaling pathway is also one of the important signaling pathways in eukaryotic cells. In Saccharomyces cerevisiae, ScVAC14and ScVAC7are involved in the process of phosphorylation of phosphatidylinositol. RAS-cAMP signal pathway in Saccharomyces cerevisiae plays an important role in cellular perception of nutrient substances, metabolism, proliferation, differentiation, stress resistance and mycelia growth, and these processes described above also play important roles in Magnaporthe oryzae pathogenic mechanism. However, little knowledge on the homologous genes of VAC14, VAC7and IRA1in M.oryzae development and pathogenesis has been obtained and a further reseach is required.In this study, I tried to clone and knock out Molscl and MoPlbl, and analyze the phenotype of the△vac14、△vac7、△iral mutants. The results as follow:the null mutant of Molscl was obtained, however, its pathogenicity didn’t change. The phenotype of the mutant△vac14was different from the wild type, especially on the pathogenicity, meanwhile the mycelium of the mutant△vac14,△vac7were abnormal. For mutant△iral, its conidiation increased, and the germination and appressorium formation of△iral accelerated.
