In order to study the30%energy restriction in broiler spleen and bursa differentgene expression, this experiment chose of48-day-old healthy female AA broilerspleen and bursa tissue(energy restriction and freedom group),each group20samples,a single individual for the repeat units. we used GenefishingTM DEG Premix Kit toselect spleen’s and bursa’s differentially expressed genes, used semi-quantitativeRT-PCR for the identification of differentially expressed genes. Following were thetest results:(1) Through the GenefishingTM RT-PCR,we detected33differentially expressedgenes in broiler spleen, in which16differentially expressed fragment showed aupward trend, a downward trend for17,while2differentially expressed fragmentwere specific expression whose were named respectively the P4-2and P12-4, P4-2showed specific expression in freedom group, the expression of P12-4was specific inenergy restricted group.21differentially expressed EST were successfuliy recovery,cloning and sequencing in broiler spleen. In broiler bursa, we detected40differentially expressed genes, in which27differentially expressed fragment showeda upward trend, a downward trend for13, while1differentially expressed fragmentshowed specific expression in energy restricted group which was named F8-2,17differentially expressed EST were successfuliy recovery, cloning and sequencing(2) Through semi-quantitative RT-PCR we showed that:9of13differentiallyexpressed EST were significant differences, and the others were no significantdifference in broiler spleen.7of11differentially expressed EST showed significantdifferences, the other were no significant difference in broiler bursa.(3) Through bioinformatics analysis we showed that:4differentially expressed ESTcan be found highly homologous known genes,4are speculated genes, the othergenes function is unknown in broiler spleen. In broiler bursa4differentially expressedEST can be found highly homologous known genes,2are speculate genes, the other genes function is unknown.In this study, We screened and identified differences in energy restricted broilerspleen and bursa displayed EST.This provided a reference for human researchingenergy restriction effects in broiler from the molecular level.
