| The impact of gibberellin and cytokinin on seed germination and seedling growth of South Korea and FuSong Damaya ginseng were studied. And the difference of germination rate, seedling growth and plant form between two different genetic ginsengs was analysis to reveal the forming mode of ginseng aboveground which mediated with cytokinins. And the technological method of ginseng "muti-stem" forming would be studied with this theory basis clarifying. It also have advantage for the field work of control the growing development of ginseng.The impact of different concentration gibberellins on splitting rate of ginseng seed from South Korea and FuSong was studied. The results showed that the seed after-ripening time was shorten, the breach percentage was increased with remarkable concentration effects, and the best treatment was200mg/L. Both the South Korea and FuSong seed were basically coincident with the law of ginseng seed after-ripening of seed-physiology.The influence of after-ripening time of seed-physiology, cytokinins concentration and soaking time on ginseng seed germination, seedling growth was examined. Orthogonal experiment was designed and analysis with5indexes, including germination rate, seedling stem length, root length, fresh weight of whole plant and leaf. The result showed that (1) the effective treatment combination for seed germination and seedling growth of South Korea ginseng was:60d of cold treatment,20mg/L of6-BA, immersion time was24h. And the cytokinins concentration showed a more obvious influence than other2factors.(2)the effective treatment combination for seed germination and seedling growth of FuSong ginseng was:60d of cold treatment,20mg/L of6-BA, immersion time was24h. And the effect of after-ripening time, cytokinins concentration on seed was more remarkable than soaking time.The impact of cytokinins on quantity of leaf and petiole of seedling, proportion of nastic movement changes, germination of lateral bud was studied, and different parts were taken photos or made paraffin section respectively. It was found that the South Korea ginseng seedling was more sensitive to cytokinins. The main changes included (1) the quantity of petiole and leaf was increased with the emergence of a double petiole, and2,4and5pieces of leaves (3pieces of normal).(2)The phenomenon of spiral growth and lateral growth of stem were observed, which meant the geotropism of seedling was changed. And the treatment with highest proportion(80%) of variation was the seeds when cold storage for30d.(3) Germination percentage of lateral bud was increased, the treatment of20mg/L6-BA showed the best effect(52.9%). |
