Objective:To understand the influence of the graded seedlings on the yield and quality of medical material, the connection between the size of root and the breeding seedlings, and establish the quality classification standard of5. miltiorrhiza Bunge. To explain the rooting mechanism of S. miltiorrhiza Bunge. by studing the physiological-biochemistry variation of root segments during taking roots and buds.Methods:S. miltiorrhiza Bunge seedlings were graded primarily, planted, and its growth situation, such as plant height, number of inflorescence and branches, the yield and quality of annual and biennial were observed and compared.The research of seedlings bred with root segments were finished in field. The root segments were divided into8groups according to length, diameter and position, and the morphologic changes of seedlings were surveyed, the date were analysed by K-means cluster, and divided into3classes. The soluble sugar、soluble protein and ammonia nitrogen in root segments were determined by UV spectrophotometer in rooting mechanism research of S. miltiorrhiza Bunge, and the endogenous hormone and chemical constitution were determined by high performance liquid chromatography(HPLC).Results:①The coefficient variation of S. miltiorrhiza Bunge seedlings’ character was great, and a year later there were differences in the yield and the level effective component content of S. miltiorrhiza Bunge between different groups, with the effective component content meeting Pharmacopoeia requirements. The yield of biennial was sharply increased compared to the annual, but the effective component content was lower than the annual. The order of the tanshinones content and the classification of the annual showed a basically opposite trend.②The seedlings bred with the longer root segments had higher fraction surviving, higher plant height and less inflorescence. There were difference in plant height between different length groups, and the primary interaction between root diameter and position made a difference of plant height;the secondary interaction between length, diameter and position had an effect on number of branches;the number of root branches between different diameter groups and position groups was different; and the dry weight and root diameter were different between different position groups.③The soluble sugar content in three groups showed a "M" trend, the soluble protein content was stable in whole period, ammonia nitrogen content fell down in prior period, then slowly increased. Fatsoluble substance content varied with similar downtrend. During taking root and budding, only auxin content increased, the ratio auxin to abscisic acid was high.Conclusion:The S. miltiorrhiza Bunge seedlings should be graded, and the quality of the annual was better than the biennial, the yield of which was less than the yield of the annual. S. miltiorrhiza Bunge is easy to take root, the longer, thicker and upper segments, the higher fraction surviving, earlier rooting and budding and greater weight. During taking root and budding, the content of primary and secondary metabolites and endogenous hormone varied.
