In addition to classical antibodies IgG1,there is a significant amount of unusual (with simplified structure) non-canonicalantibodies IgG2and IgG3in blood of Camelus Bactrianus. And their content up toabout60–80%. Due to their unique structure, make its have many kinds of specialbiological function. In order to observe characteristics of distribution of the plasmacells secrete IgGs and reveal the relationship between plasma cells secrete IgGs andmucosa immunization. The details of study were divided into some parts as follows:8Camelus Bactrianus’ serum were collected. The protein was separated throughammonium sulfate precipitation of different degree of saturation, which was started20%of saturation and With5%of the gradient gradually increased to20%ofsaturation. Each precipitation with centrifugal experiment respectively. According tothe content, molecular weight and structure characteristics of IgGs of the CamelusBactrianus’ serum, to make predictions and certain about the conditions of maincoprecipitations of IgGs of the Camelus Bactrianus’ serum though the proteinelectrophoresis of SDS polyacrylamide gel. The results shown that the content ofIgGs of the Camelus Bactrianus’ serum was the largest by35%~40%ammoniumsulphate precipitating after dilute it with equal amount of saline water. It is the mianrange about the precipitates of IgGs.Sediments of the35%saturated(NH4)2SO4were purified by the Sephadex G-200gel chromatography column and DEAE-52ion exchange chromatography columnwhich used in combination according to Camelus Bactrianus blood serums IgGsmolecules of physical and chemical properties. The contain of protein were detectedby ultraviolet spectrophotometry, and curve drawing and analysis elution curve. The results showed that two kinds of purification methods used alternately, complementeach other, finally got of high purity Camelus Bactrianus serum IgGs.Mixed immune method about injection of multi-point into dorsal subcutaneoustissues, lymphonodusand the ear vein was used to the health of adult rabbits.Antibodytiter was detected by plateagar immune double diffusion test. The resultsshowed that Antibodytiter was1:512at last and the expected objects were achieved.The blood of rabbits were collected through their hearts. And the rabbit polyclonalantibody were purified.It was observed that the characteristics of distribution for the plasma cells of CamelusBactrianus serum IgGs in the small intestine and statistical analysis them. though usedthe methods about anatomy and immunohistochemistry. The results showed that theplasma cells of IgGs were mainly distributed in the base of intestinal villus in theduodenum, which dispersedly existed in it and around the intestinal gland were more.Some of them crowded around it. Occasionally, it were observed in the brunner’sgland. It is not discovered in the enterocyte, lymphoid follicle, smooth muscle andserosa. The characteristics of distributions are similar which are in the jejunum andthe ileum (except the brunner’s gland). The statistical results showed that the densityof the plasma cells of IgGs were reduced gradually from front duodenal to terminalileum, which were in the lamina propria. Experimental results proved that the plasmacells of IgGs mainly existed in the small intestine of Camelus Bactrianus, the densityof them were reduced gradually from front duodenal to terminal ileum. It seems thatthe relationship of the characteristics of distribution is positive with respect to theabsorption of nutrients in the small intestine of Camelus Bactrianus.
