The yak blood utilization rate is relatively low, at the same time the content ofhemoglobin in the blood is higher, the heme has high edible and medical value, so theextraction of heme from yak blood has a high practical value. In this study, the yak blood asthe raw material were used in methods of CMC-Na, acid acetone and enzyme to research theheme extracting process and the optimal parameters. Comparison of different methods ofextracting heme from yak blood, find out the characteristics of each method, as well as in yakblood heme extraction application value. The results showed that:1. Orthogonal test was carried out according to the single factor test results and table L9(3~4). The bestextraction process of the yak blood heme by the method of CMC-Na as follows:100ml water,ultrasonictime is15min,pH value is2, CMCNa (3%) adding quantity is8ml.The result show that heme purity is10.16%, the extraction yield of yak blood heme is3.45mg/ml.2. According to the single factor test results, orthogonal test was carried out according totable L9(3~4). The acidic acetone optimum extraction process as follows20ml yak blood,30mlwater, ultrasonic time was15min,100ml acetone, pH value is2.5. The result show that hemepurity is14.37%, the extraction yield of yak blood heme is2.63mg/ml.3. Using the single factor test results and L9(3~4) orthogonal test, the enzymatic extractionof yak hemoglobin process was optimized as follows:20ml yak blood,100ml water,ultrasonic time is10min, pH value is3, enzymolysis temperature is50℃.The result showthat heme yields is1.1085g, the purity is13.64%, the extraction yield of yak blood heme is3.40mg/ml.4. Compared the results of the three methods, CMC-Na method has characteristics ofhigh extraction rate, low production cost and simple production process. Acidic acetonemethod, has high production cost, low extracting rate and high purity. Enzymatic method hashigh extraction rate, low production cost and complicated process.As far as the cost of production, CMC-Na and enzymatic method are far less than acidacetone method.They can be used as a good and practical extraction methods. But from theproduction perspective, using the method of CMC-Na extraction yield was significantlyhigher than that of acidic acetone method and enzyme method.It can be used as low purity ofheme extraction method. In the application of High purity heme,second extraction purity size: acidic>acetone>CMC-Na,and the purity of their production were difference, theywere86.16%,54.87%,29.59%.
