Waste beer yeast mud have a lot of active elements,waste beer yeast has beenused to the feed through drum drying methods by brewery at present.It have moreproperty of nutrition and medicine.The beer of China output is490.2millionton.Calculate in advance,Compare with traditional fermentation and beermanufacture.The more beer output enhance every year,The more waste beer yeast willbe produce in future.So from waste beer yeast extract effective component is morefeasible and significance.Waste beer yeast have a kind of enzymes is superoxide dismutase (sod),the EMShas forbit use the SOD from cattle blood in1997.The main originate of SOD is plantsand microorganism at present.The amounts of plant has less than blood.But beer havea lot of SOD. So this way can better develop and utilize the functional elements in thewaste beer yeast powder.The product have high safely function.waste beer yeast has been used to the feed through drum drying methods bybrewery at present. Most manufacturer use the waste beer yeast add to feed by theroll dry way.Because of instrument rough and raw material,some product have morewater member.And some product has been undergo heated too much time,nutritionhas been destroied.Even produce harmful product.It will effect secure.Usually,SOD can against for heat about sort time.the resistant ability isperformance. This method can protect the functional elements in waste beer yeast notto be destroyed and it is easily to store up and transport.The main research contents and results are as follows:1.Through the spray drying principle of work and the actual operation processselection influence the spray drying process of the main factors, the influence of thespray drying process parameters have feed concentration, feed rate, into the air, inletair temperature, etc.Determine the feed rate, into the air, the optimization of the inlet air temperaturerange of response surface center fitting optimization. According to the actual situationof production and demand, this study selected superoxide dismutase SOD activity forinspection index, coomassie brilliant blue method measuring protein content andimproved pyrogallol oxidation test since SOD activity. Get feed concentration20%,the feed rate of250mL/h, the inlet air temperature of120℃, the SOD activity of airvolume of80m3/min is better. 2.Through the single factor to determine the parameters of optimized parametersand, according to the actual situation of spray drying, feed concentration choose25%,According to the quadratic regression equation of the spray drying optimized wastebeer yeast mud optimum parameters for inlet air temperature134℃, feed rate of225ml/h, into the air72m3/min. The experimental data validated that the best parameterequation in error allowed range, accord with practical significance.3.Extracting SOD from waste beer yeast powder by the ultrasonic-microwaveassisted.As the single factor to determine the suitable condition,to be select salt(NaCl), ultrasonic intensity, ultrasonic action time, microwave action time for brokenwall of the influence factors to SOD vigor for testing factors. This study adoptsultrasonic and microwave-assist method to extract SOD to optimize the bestextraction parameters for ultrasonic intensity574w, ultrasonic time10.85min,microwave time2.7min, add salt3%. To predict model verification test, the results ofthe validation of the model proposed, do not break the permissible error range, haspractical significance.4.Determined by the experiment ultrasonic-microwave assisted extractionmethod of waste beer yeast powder of SOD process: SOD dry yeast powder1g-NaCl solution and ml soak12h,4℃and ultrasonic oscillations574w,10.85min,low temperature microwave heating2.7min to4000r/min, centrifugal3min-toprecipitation, leave supernatant fluid, adding suitable amount of cold acetone, shockafter the uniform standing for30min to4000r/min centrifugal5min andprecipitation in distilled water of SOD crude enzyme solution.5.Separation the sterling SOD by the Sephadex G-75chromatography column.Through the contrast effect Sephadex G-75chromatography column of the mainfactors, appropriate separation conditions, and through the ultravioletspectrophotometer spectral scanning and SDS polyacrylamide electrophoresis topurified enzyme solution for purity detection. Get the sample quantity1ml, chooseTris-HCl buffer as eluent. Keep1ml/min flow better effect.6.The results get the spray drying processing waste beer yeast mud optimumprocess parameters, get low temperature microwave assisted extraction method ofwaste beer yeast mud SOD the best process. Use the spray drying technology insteadof drum drying technology processing waste beer yeast mud, save the functionalcomponents, with low temperature microwave assisted extraction method of wastebeer yeast mud of SOD.
