As efficient tool in organic synthesis, enzymatic synthesis has been development rapidly. In recent years, more and more chemists focus on the research of the enzymatic multicomponent synthesis, and provides new ways for the green catalytic.Heterocyclic compounds in organic chemistry is one of the popular research field. Heterocyclic compounds which contain thiazole have good pharmacological and biological activity. It plays an important role in the chemical, pharmaceutical and pesticde and have important research value.In this thesis, we studied the methods of the enzymatic synthesis.Part1:Three component synthesis of2,5-substituted thiazole derivatives catalyzed by Typsin from porcine pancreas. We screened8commercial enzymes to catalyzed the reaction of secondary amines, benzoyl isothiocyanate. and dialkyl acetylenedicarboxylates, we found Typsin from porcine pancreasshowed the highest catalytic activity in the reaction. Experiment investigated influence of different factor including solvents, enzyme sources, tempratures, reaction time and enzyme concentration in the enzymatic reaction, and112,5-substituted thiazole derivatives were prepared and we proposed a possible mechanism for the reaction.Part2:Three component synthesis of4-thiazolidinones derivatives catalyzed by Typsin from porcine pancreas. We investigating solvents, enzyme sources, tempratures, reaction time, enzyme concentration and the effect of structure of substrate, and204-thiazolidinones derivatives were prepared under optimized conditions.Part3:Four component synthesis of2,4-substituted thiazole derivatives catalyzed by Lipase from Porcine pancreas. We screened the different commercial enzymes to catalyzed the reaction of amines, aldehyde, thioacetic acid and methyl3-(dimethylamino)-2-isocyanoacrylate, we found Lipase from Porcine pancreas showed the highest catalytic activity in the reaction. We confirmed the quaternary structure of lipase from rorcine pancreas play a decisive role in this reaction and152,4-substituted thiazole derivatives were synthesized. It shows that the enzyme has good universality in this reaction.The thesis provides three methods to synthesis the heterocyclic compounds which contain N,S, and we obtained46compounds. The products were characterized by IR、GC-MS、1H-NMR、13C-NMR.
