| Crude polysaccharides were extracted from seed-watermelon flesh by Water extraction and alcohol precipitation. Meanwhile, effects of material-liquid ratio、 temperature, pH value, and diffusion time on polysaccharides extraction ratio were studied through orthogonal experiments L9(33) in order to raise the polysac charides extraction ratio. Finally, the optimum conditions of polysaccharides extraction were: material-liquid ratio1:100, temperature95℃, diffusion time3.5h, and the extraction ratio was2.803%when the extracting times was1.The crude polysaccharide was fractioned by Sevage law to take off the protein, then separated and dialyzed by Sephadex G-75gel filtration, the monosaccharide components can be obtained by efficient ion chromatography from polysaccharide component, the results are:rhamnose、arabinose、galactose、glucose、xylose、galacturonic acid、 glucuronic acid, The content of various components:rhamnose8.12%, arabinose10.7%, galactose63.96%,glucose5.43%,xylos7.14%,galacturonicacid4.22%,glucuronic acid0.43%。The Antioxidant activity results of extracting polysaccharide isolation from melon seeds matures showed that:The most clearance rates for·OH of the melon seed matures polysaccharide was28.91%, meanwhile the maximum Inhibition rates for MDA was10.46%, and the most clearance rates for·O2-was15.17%. In the same concentration conditions, The clearance rates for·OH and·O2-and the inhibition rates for DNAs of the melon seed matures polysaccharides were lower than Vc.The function of melon seed matures polysaccharide to reduce blood high blood fat were evaluated, the results showed:High doses melon seed matures polysaccharides could reduce the weight and Spleen index of high blood fat mice, meanwhile increase thymus index.
