There are abundant vitamins, flavonoids, mineral elements and other natural active substances inhippophae rhamnoides, which are mainly existed in its roots, stems, leaves and fruits. Because theflavonoids have many kinds of valuable function, such as antioxidant, the hypoglycemic effects and so on, inthis paper, the research object of this paper is the flavonoids substances. And also considering the exploitationof hippophae rhamnoides fruit has been reported a lot, the comprehensive utilization of hippophaerhamnoides plants is our new ideas. The research is focused on the flavonoids extraction from the leaves,purification and the mice in vivo experiments.1. At first, ethanol was the optimum entrainer was determined. The effect of ultrasonic power, extractiontemperature, extraction time, ratio of materials to liquid, the concentraction of ethanol on the efficiency offlavonoids from hippophae rhamnoides leaves were studied. The quadratic general spinning design and SASstatistics analysis method were adopted to optimized the process of extraction flavonoids from hippophaerhamnoides leaves. The results showed that the optimum extraction conditions were as follow: extractiontime31min, the concentration of ethanol was76%, the ultrasonic power was562W, the ratio of materials toliquid1:36, under the conditions of the flavonoids of the hippophae rhamnoides levaes was10.51mg/g.2. Comparing the efficiency of absorption and elution of D3520,X-5and AB-8to flavonoids ofhippophae rhamnoides leaves, X-5adsoptive resin was optimum. The static parameters of abosoption andelution were: absorption time3h, absorption temperature25℃, elution time2.5h, elution temperature25℃,ethanol concentration65%. Studying the condition of dynamic absoption and elution of X-5resin, theoptimum parameters of the absoption capacity2.5mg/ml, abroption rate2BV/h were determined. Theoptimum elution parameters were ethanol concentration75%, elution rate2BV/h, elution capacity3.0BV.The purity of flavonoids of hippophae rhamnoides leaves was89.72%under the above conditions.3. This study adopts the high fat diet mouse model to evaluate the pharmacological activity of the totalflavonoids of Hippophae rhamnoides leaf lipid-lowering. Fed high fat diet of mice body weight per week is asubstantial increase, there is always a significant difference (P <0.01) compared with normal control group,model group and normal group of mice4weeks after the lipid water also exist significant difference (P<0.01), the simultaneous administration group and the high dose group from the weight and serum lipids andliver index and the normal control group differences, with the positive control group, significant differences(P <0.01), medium dose group and positive group differences significant (P <0.05), low-dose group changedlittle slice observation of the internal organs of mice after the administration of the mouse visceral lesions, theextraction of this experiment the material is safe and nontoxic.
