The essay research was studying collagen polypeptide power from Navodon septentionalis fishy goods factories’ bone waste, using biotechnological enzyme engineering technology through optimizing protein enzymes and controlling the process of hydrolysis, to make Navodon septentionalis collagen polypeptide and analyse of its antioxidant properties and indicators of producing.Through the small and medium producing process.the bone collagen power was made out to improve the basis of flavor products or other funtional products.Firstly, the basis producing funtional indicators were determined through the determination of protein.fat and water content of Navodon septentionalis bone including fishhead, skaped, main fish bone.With high pressure preparation ways to extract some collagen popypeptide from bone. its basic biochemical characters were also determined.Secondly, the appropricate protease and conditions of hydrolysis were selected out to gain biochemical and high content collagen polypeptide with Response Surface Methodology. The high biochemical activity collagen peptide was prepared under the following conditions that was bone collagen was hydrolyzed at natural pH(about5.6) with flavor protease6.92%o and water content42.27%and keeping reacting2hours at55℃.The content of collagen peptide was about1.29mg/ml. Through Ultraviolet detemined analysis, the biggest absorption of collagen polypeptide is about220nm; with Fourier transform infrared spectrum analysis, it has strong absorption peak at1646cm-1and1570cm-1with the similar functional structure of other fish skin or bone collagen polypeptide;by analysis of high performance liquid chromatography.the total amino acid is up to77g in100g polypeptide power and the content of hydroxyproline is about3%s;with the determination of MALDI-TOF-MS.the molecular weight of collagen polypeptide concentrates is below3000Da.At last, the antioxidant activities of different prepared methods’collagen polypeptide were determined through the reduction power, scavenging properties on hydroxy radical and DPPH compared with GSH on different contents. The reducing power of bone collagen peptide at100mg/ml was0.833, the IC50of scavening hydroxy radical and DPPH radical are21.5mg/ml and15mg/ml,respectively.
