Endophyte is a microorganism community with abundant biodiversity. Their existence and effect on the host plant have been ignored for the reason that they lived in the healthy plant tissues without causing any diagnostic and co-evolved with their hosts. With the development of endophyte research of medicinal plants, bioactive compound from endophyte has become a new hotspot.The present paper has studied the Catharanthus roseu, Cephalotaxus hainanensis and Antiaris toxicaria endophytes of three medicinal plants with the anti-tumor activity in Hainan Province. A further study was needed for the strong activity of strain PF09005. The results were as follows.204 strains of endophytes were isolated from C.roseu, C.hainanensis and A.toxicaria and their anti-tumor activity of fermentation samples were pre-screened by the method of MTT. The results showed that 19 strains presented cytotoxic activities to at least one tested tumor cells, accounting for 9.31%. Among them strain PA09006, PA09008, PA09009, and PF09005 possessed prominent bioactivity and genetic stabilization.By studying the morphology, cultural characteristics.Strain PF09005 was identified as Arthrobotrys dactyloides.The optimal liquid fermentation conditions and media of PF09005 were found out. The optimum culture media of PF09005 contained 20%potato, glucose 2.0%, sucrose 2.0%, yeast extract 1.0%, peptone 0.5%. The optimum fermentation conditions of PF09005 were initial pH7.0, the size 200mL in 500mL erlenmeyer flask at 180 r·min-1 and 28℃for 9d. Under the optimum conditions, the inhibitory rate of S180, increased from 83.2%to 96.1%.The physical and chemical properties of the anti-tumor compounds from PF09005 have been studied, it is demonstrated that the bioactive compounds were relatively stable under the room temperature and even in warm-water bath, and the pH value ranging from 5.0 to 8.0, however, unstable under the UV-light or long time sunlight condition. They can be extracted in chloroform, ethyl acetate, n-Butanol.
