Because of its inherent characteristics yeast cells are always used for scientificresearch。 Saccharomyces cerevisiae can switch between the haploid and diploidforms, and is used as a eukaryotic model organism for scientific research. Thereversible phosphorylation of proteins in eukaryotes is a key regulatory mechanism incellular signal transduction pathways. Protein kinases and protein phosphatasesregulate the phosphorylation state of their substrates in order to achieve theintracellular signaling pathway activation or inactivation, thereby affecting thereproductive, differentiation, morphological transformation and apoptosis.Protein phosphatases are divided into three subfamilies: protein tyrosinephosphatase, protein serine/threonine phosphatases and dual-specificity proteinphosphatases. One of the protein serine/threonine phosphatase members, type2Cprotein phosphatases are monoenzymes, and their activity is dependent on themagnesium ion or manganese ion. Until now, there are seven PP2C proteinphosphatases identified in S. cerevisiae, including ScPtc1p, ScPtc2p, ScPtc3p,ScPtc4p, ScPtc5p, ScPtc6p and ScPtc7p, so are in C. albicans, CaPtc1p, CaPtc2p,CaPtc4p, CaPtc5p, CaPtc6p, CaPtc7p and CaPtc8p. In both Saccharomycescerevisiae and C. albicans, Ptc5p, Ptc6p and Ptc7p are all located in themitochondrion.In C. albicans, deletion of PTC5, PTC6or PTC7does not show anyphenotype. To further study the function of these genes, we constructed C.albicansptc5ptc6and ptc5ptc7double mutants as well as the triple-gene mutant ptc5ptc6ptc7.Phenotype analysis indicates that there is a genetic interaction between C. albic PTC6and PTC7in azole sensitivity.
