Objective:1. To establish an appropriate way of feeding, breeding for daurian ground squirrel (Spermophilus dauricus) in laboratory and investigate the rules of its hibernation bouts.2. To compare the changes of regulation of cytoplasmiccalcium of skeletal muscle and its sarcoplasmic reticulum of daurian ground squrriels and rats.3. To study the effects of Ligustrazine on regulation of cytoplasmic calcium of skeletal muscle and its sarcoplasmic reticulum of tail-suspension rats.Methods:1. The hibernation room was made according to the parameters of wild daurian ground squirrel’s caves in the field and the hibernation bouts were monitored by the traditional sawdust method.2. The activity of Na+,K+-ATPase and Ca2+-ATPase of skeletal muscle and SERCA was tested by determination of inorganic phosphorus.3. The expression of SERCA2isoform was tested by Western-Blotting.Results:1. The survival rate of animals was66.7%, and survival rate of breeding body was78.9%, the biggest relative fatnesses is4.04g/cm3Autumn, increased34.15%(P<0.01)、50.88%(P<0.01) and21.46%(P<0.01) compared with SPR、SUM and WIN group.2. The average time of hibernation is93.95d in the hibernated house. The average days of each hibernation bout was7.44and the arousal days of hibernation bout was1.36. The days of occurrence of hibernation bouts accounted for89.9%of all days. There were7.55hibernation bouts of animals during hibernation.3. Compared with CON group, Na+,K+-ATPase, Ca2+-ATPase of SOL of TS+W group increased46.99%(P<0.01) and38.57%(P<0.01); Na+,K+-ATPase, Ca2+-ATPase of SOL of TS+TMP group increased25.36%(P<0.05) and21.20% (P<0.05).Compared with TS+W. Na+,K+-ATPase of SOL of TS+TMP group increased14.71%(P<0.05).There was no significant difference of Na+,K+-ATPase and Ca+-ATPase of EDL of rats.4. There was no significant difference of Na+,K+-ATPase and Ca+-ATPase of SOL and EDL of daurian ground squirrels in tail-suspension.There was no significant difference of Na+,K+-ATPase and Ca2+-ATPase of SOL among Pre-H,H and Post-H group.The lowest of Na+,K+-ATPase and Ca2+-ATPase of SOLwas IB-A group.and the highest was Next-H group. There was no significant difference of Na+,K+-ATPase and Ca2+-ATPase of EDL among Pre-H,H, IB-A and Next-H group.and the hightest was Post-H group.5. Compared with CON group, SR Ca2+-ATPase of SOL of TS+W and TS+TMP increased30.55%(P<0.05) and25.78%(P<0.05) and SERCA2increased21.79%(P<0.01) and20.90%(P<0.05). There was no significant difference of SR Ca2+-ATPase and SERCA2between TS-W and TS-TMP group.There was no significant difference of SR Ca2+-ATPase and SERCA2of EDL in rats.6. There was no significant difference of SR Ca2+-ATPase and SERCA2of SOL and EDL of daurian ground squirrels in tail-suspension. Compared with AUT group,SERCA2of SOL of AUT-TS increased64.94%(P<0.01).while EDL had no difference. There was significant difference of SR Ca2+-ATPase and SERCA2of SOL among every group and there was no difference of SR Ca2+-ATPase in EDL.SERCA2of EDL of IB-A and Next-H group increased compared with other group.Conclusion:1. Wild daurian ground squirrels can be successfully bred and fed in lab condition and achieve their hibernating period in artificial hibernation room.2. The activity of Na+,K+-ATPase and Ca2+-ATPase increased of tail-suspension rats.and the activity of Na+,K+-ATPase can be restrain by TMP.There was no significant effect of EDL of rats and SOL and EDL of daurian ground squirrels in tail-suspension.The activity of enzymes of cytoplasmic regulation decreased in hibernation.3. The activity of SR Ca2+-ATPase and SERCA2of SOL of tail-suspension rats increased,and there was no significant change of EDL.The SERCA2of SOL of daurian ground squirrels increased in tail-suspension,but the activity of SR Ca2+-ATPase didn’t. The activity of SR Ca2+-ATPase and SERCA2of SOL of daurian ground squirrels increased in hibernation,and there was no significant change of EDL.
