| Extracting method of tea saponin from the cakes of camellia oleifera which has been extracted tea oil by combining ultrasonic was introduced.And AB-8macroresin was used to purify the tea saponin samples.We tested its antioxidation ability and antibacterium ability.The saponins and monosaccaride were obtained by acid hydrolysis process.TLC was employed to analyse the constituent of the monosaccaride and saponins,HPLC-UV-MS was used to determine the structure of the saponins initially.Main results are as follows:1,The water and ethanol extraction process were compared with this method.The result showed that the extraction by ultrasonic-assisted ethanol extraction decreased extraction time by about7h.The optimum extracting process was selected by the response surface analysis,the optimum parameters are:ultrasonic power800W,frequency20kHz, extraction temperature is60℃,extraction time is30min,the ratio of sample to solution is1:10,ethanol concentration is70%.,two times extration and the final yield reach89.9667±0.1429%.2,We used AB-8macroresin to purify the tea saponins samples.Its dynamic and static adsorption ratio were0.0356g/g dried resin,94.15%.At the condition of solution concentration10mg/mL,PH6.3,flow rate2BV/h,the AB-8macroresin showed the better comprehensive property.Then the following technological condition:distilled water,0.2%NaOH,25%,45%,65%,85%,95%ethanol as eluting reagent,gather the part eluted by65%ethanol,and the purity is80.56%after HPLC ananlyzing.3,The purified tea saponins were acid hydrolysis with10%HCL solution for5h,then the saponins and digest solution were obtained.TCL were used to analyse the saponins and monosaccharides.The result insisted that only glucose exist in the tea saponins and the saponins included two different components.4,HPLC-UV-MS detected the sapoins,we found that after the acid hydrolysis,there were three isomerides,their molecular mass were about1521.IR and mass spectrometry were applied to analyse the structure of saponins. 5,The inhibiting ability of tea saponins were certained by a series of experiments such as reducing power determination,eliminating the superoxide anion radical in the Tris system ability determination,eliminating the hydroxyl radical ability and DPPH and so on.After the bacteriostasis experiment of Staphylococcus aureus,Colon bacillus,Bacillus subtilis,the results inferred that tea saponins solution has the bacteriostasis ability to this three microorganism.When the concentration is5mg/mL,tea saponins can inhibit Staphylococcus aureus and Colon bacillus,the Bacillus subtilis only could be inhibited when the concentration is10mg/mL.Usually,the tea cake extrated tea oil before were discarded or used as feed for abranchiate.There are10%-13%tea saponins in the tea cake,and tea saponins are a very useful component. So,it's meaningful for increasing the rate of multipurpose utilization and its value when we use the remaining tea cake as the material of tea saponins extration.Compared to traditional tea saponins extration,ultrasonic-assisted tea saponins extration is more time-saving,lower energy consumption and higher yield of tea saponins. |
