The Effect Of Chinese Medicine Tian You Capsule On Anti-Addiction

Drug addiction is a chronic relapsing disorder which is characterized bycompulsive drug seeking and drug taking behavior despite serious adverseconsequences and which is caused by long-term use of addictive drugs. Drugabuse has become a main medium of the severity of infectious diseases suchas AIDS and hepatitis, at the same time the violent crime caused by drugabuse seriously harm human health and affect social stability. Moreover,addicttion relapse rate is as high as95%after treatment.Therefore, addictionhas became a social problem, and to find safe and effective anti-addictionrelapse drugs is an important task for health care workers.In China, Traditional Chinese medicine has been used for more than2,000years. In the1760s, British shipped opium to China and brought terribleimpact to China's political and economic, which was serious impediment tothe development of china. Chinese medicine began to be used for addictiontreatment at that time, and now it has been used for more than200years. Afterthe People's Republic of China was set up, the Chinese government manage toeliminate abused drug by strict law enforcement and government policyseparating China from western countries.The abused drugs almost disappearedin Chinese market, and the study of addiction treatment became less. With therecent re-establishment of the open-door policy toward western nations, Chinare-established diplomatic relations with Western countries, the drug wasre-emergenced and spread all over in China, the research on anti-addictionbecame popular again. The advantages of Chinese medicine on addictiontreatment are it can prevent relapse by effectively reduce withdrawal andresidual withdrawal Symptom with little side effects. Therefore in recent years, traditional Chinese medicine became more and more popular used for drugaddiction treatment. A great many of results have been achieved through yearsof research, and a lot of chinese medicine compounds has been produced, butChinese medicine research mainly focuse on anti-withdrawal symptoms andanti-residual withdrawal symptom, the treatment to prevent relapse inaddiction, the most important aspects of anti-addiction research,is studied verylittle.Objective: This study aims to study the effect of traditional Chinesemedicine Tian You capsules on anti-addiction, and expect to develop a highefficiency and low toxicity medicine to anti-addiction relapse.Methods: Our investigation adopt classic model of addiction, such asConditioned Place Preference, Behavioral Sensitization and Self-administration model,to evaluate the effect of Tian You capsules onanti-addiction relapse.Results: The research of this study is discovered as follows:(1) The effect of Tian You capsules on positive reinforcement effect ofaddictive drugs1) The experiment of conditional place preference induced by morphinein mice (Conditioned Place Preference, CPP): First of all, Tian You capsules(6.25,12.5,25,50,100mg/kg) does not induced conditioned placepreference, which suggest that Tian You capsules does not induce positivereinforcement effect; Second, compared with the morphine control group(Mor group), TianYou capsules combined with morphine conditioning failedto affect the acquisition of morphine-induced CPP; Finally, acuteadministration of Tian You capsules (12.5,25,50,100mg/kg) does not affectthe expression of morphine-induced CPP in mice.2) The experiment of morphine induced behavioral sensitization inmice:TianYou capsules (12.5,25,50and100mg/kg) combined with morphinedid not affect development of morphine induced Behavioral Sensitization.3) The experiment of heroin-induced self-administration in rats: First, underfixed-ratio2(FR2) program, Tian You capsules (120,150,180,210mg/kg) does not inhibit the development of heroin-induced self-administration in rats;Second, acute administration of Tian You capsules (120,150,180,210mg/kg)does not affect the drug taking behaviors in rats; Finally, under FR5program,the acute administration of Tian You capsules (180,210mg/kg) also does notaffect the self-administration behaviors.In summary, Tian You capsules has no suppressive effects on positivereinforcement of opioid drug, and thus we study the treatment of relapseprevention, which is the most important part of the addiction treatment.(2)The study of Tian You capsules in relapse prevention1) The experiment of CPP reinstatement induced by morphine in mice:Treatment of TianYou capsules (25,50and100mg/kg, i.g. tid) in the sessionof extinction significantly decrease morphine-primed reinstatement of CPP.2) The experiment of expression of behavioral sensitization induced bymorphine in mice: TianYou capsules (50and100mg/kg, i.g.) prior toexpression of Behavioral Sensitization significantly block the expression ofmorphine induced Behavioral Sensitization.3) The experiment of the extinction of self-administration: chronicadministration of Tian You capsules (150,180mg/kg) significantly weakdrug-seeking behavior in addicted rats4) The experiments of heroin-induced self-administration relapse in rats:Acute administration of TianYou capsules (180,210mg/kg) inhibit heroin(250μg/kg) induced ralapse of self-administration.In summary, TianYou capsules can effectively inhibit relapse in animals,but whether this function of block drug addiction has a particularity role, weproceeded spontaneous activity experiments.(3)The effect of Tian You capsules on the spontaneous locomotor activityFirst, the acute administration of Tian You capsules(12.5,25,50,100mg/kg)significantly inhibit the spontaneous activity from the20'th minute to the30'thminute after administration of high doses of100mg/kg and then it returned tonormal after40minutes, but in the90-minute test, the distance of the totalactivity did not change; Secondly, Tian You capsules (12.5,25,50,100mg/kg) were administrated for14days, On day7and day14, after administration ofTian You capsules100mg/kg, spontaneous locomotor activity wassignificantly inhibited between the20'th minute and the30'th minute; after15days of long-term administration of Tian You capsules, spontaneous activityhas no significant different between each group without Tian You capsules.Conclusion:Our results show that Tian You capsules itself can not causeaddiction and affect the formation and development of addiction, but Tian Youcapsules can effectively suppress relapse. According to our findings, Tian Youcapsules is expected to be new medicine which is high efficiency and lowtoxicity to treat drug addiction.
