[Objective] The purpose of this research is to study the intervention effect of Yunnan Baiyao(YBY) to CD4+CD25+regulatory T cells (CD4+CD25+Treg) in collagen-induced arthritis rat (CIA) model[Methods] A total of80SD rats(6-8weeks old) were included.The arthritis rat(60) model was duplicated by injection0.1ml collagen emulsions in the metatarsal mat of right foot and0.2ml collagen emulsions in tail root, on the days of7th,21th after first injection,reinjected0.1ml collagen emulsion to each rat. At the days28, Animals were sacrificed.Pathological changes of synovium were examined by hematoxylin eosin (HE) stain method.TRAP stain also was used in every point to make sure that the CIA model was successful. When the model was success, they were randomly divided into3groups:Yunnan Baoyao (YBY) group, CIA group, Methotrexate (MTX) group, the control group was used the normal rats.There are20rats in every group.The following every4week, YBY group and MTX group were administrated by Yunnan Baiyao and Methotreate after second time reinjection, while CIA group and control group were administrated by saling with the same volumn. The rats were killed at intervals of one week. We detected the level of CD4+CD25+Treg by the flow cytometry, observed the inflammation tissue by hematoxylin eosin stain and TRAP stain, evaluated the expression of RANKLby immunohistochemical. All of this is to preliminarily discuss the intervention function and related mechanism of Yunnan Baoyao affecting CD4+CD25+Treg of spleen in rheumatiod arhritis rats[Results]The joint of CIA group appear swollen,the skin canker and dyfuction after injection collagen on day28. Hematoxylin eosin show that there were a lot of inflammatory cell in the joint.even more,the bone of the joint was destruction and erosion. Osteoclast can be detected in the bone of joint by TRAP.All appearances show that the model is successful.The level of CD4+CD25+Treg in YBY group was gradually increased at the point of week1after administrating the medication, it was stronger significantly different comparing with control group(9.57%±0.22%) and CIA group (10.84%±1.91%)(P<0.05) on weeks2. The expression of RANKL was detected at the days7with medication, but the expression went down after that point and got the same level as the control group on weeks4.[Conclusion] The model of CIA was made successfuly at the days28. The level of CD4+CD25+Treg increased after administration of Yunnan Baiyao and significant different in the weeks2, decreased gradually to normal level on the weeks4.Expression of RANKL can be reduced too in the joint organization.The results show that Yunnan baiyao may inhibit inflammation and joint bone damage absorption by raising the levels of CD4+CD25+Treg and reducing the joint organization RANKL expression. |
