The connection between innate immunity and adaptive immunity is based on the chemical mediator, chemokines and cytokines. The study on membrane molecules and cytokines of immune cells would be necessary for the further research of immno-modulation mechanisms and the treatment of diseases.In this study we managed to observe the changes in CD40 molecules in LPS-stimulated U937 cell and JURKAT cell. We used Flow cytometry to detect the expression of CD40 affter different dose ionizing radiation-and used Realtime-PCR to detect the expression of mRNA of CD40. The result illustrated that the expression of CD40 in the U937 cell is increased after the treatment of low dose radiation (LDR) and decreased after the treatment of high dose radiation (HDR), while the expression of mRNA of CD40 is increased after the treatment of both low dose radiation (LDR) and high dose radiation (HDR). But the expression of CD40 in the JURKAT cells is decreased after the treatment of low dose radiation (LDR) and increased after the treatment of high dose radiation (HDR). It was suggested that LDR might have hormesis on the immune system and HDR might inhibitite immune system.In this study we managed to construct immortalized cell model by transfecting the retrovirus vector pbabe-gfp-htert into the primary cells. First we got the htert by using restricton enzyme to digest phTERTGPlox and clone it into the retrovirus vector pbabe-gfp. Then transfect the recombination retrovirus vector into the B16 cells and used the fluorescence microscope to observe the expression of htert to confirm the expression of htert in the host cell.
