Diacylglycerol (DAG) has various physiological functions for its unique metabolic pathways. In recent years, it has been widely-used and attached more and more attention. DAG is a natural component and intermediary metabolite of oils, and its also a multifunctional additives, which has been used extensively. The thesis summarizes the physiological function, metabolic mechanism, and comprehensive evaluation of the physiological functions in rats according to procedures and methods for toxicological assessment of food by DAG as raw materials. Testes of acute toxicity and heredity toxicity, which including sperm shape abnormality test, micronucleus test of bone marrow polychromatic erythrocyte and30days feeding test were carried out on rats, then assay the influence on tissue sections of organs and changes in intestinal flora structural by denaturing gradient gel electrophoresis (DGGE) after feeding diacylglycerol in rats. Oral acute toxicity test shows that mouse oral LD50is above10000mg/(kg·bw),which suggests DAG is non-actual poison in accordance with the classification of acute toxicity. There was no influence the spindle poison and fracture effect of bone marrow cell chromosome which were fed DAG at high-dose4000mg/(kg· bw) to mouse, it means that the micronucleus test is negative. The sperm shape abnormality test within the range of tests does is also negative. Mouse after being fed DAG for30days grow well. The diet, color, action, growing development, hematology (WBC, RBC, Hb, PLT, etc) and hematology biochemistry(ALB, BUN, ALT, IP, CREA, AST, CHO, GLU, etc) index are all normal. There is no abnormal symptoms in heart, liver, kidney, spleen, duodenum and theirs'organ tissue sections after gross anatomy and histopathological examination. Then, took the PCR-DGGE analysis for colon flora in mice after30days test, the results showed that the populations multiplicity of Clostridium cluster IV and Bifidobacteria group were increased. This suggests that the DAG can have good efficacy in adjusting colon bacteria. In conclusion, DAG has no toxic side effects and plays a important tole in physiological health.
