[Objectivel1.This research is to reveal the expression and correlation of IGF-1,Caspase-3and EGFR in yong breast cancer and aged breast cancer in the way of ELSA(enzyme-linked immunosorbent assay, ELISA).2. To research the aged-related corelation about IGF-1, Caspase-3and EGFR in the patient of breast cancer.3. Analysis the role of occurrence and development about IGF-1,Caspase-3and EGFR in breast cancer and Find out the possible link from IGF-1,Caspase-3and EGFR,though the detective of the expression in IGF-1,Caspase-3and EGFR after1st3rd and5th cycles about neoadjuvant chemotherapy and judged by radiographic.[Methods]1.93cases were admitted in Third Affiliated Hospital of Kunming Medical college,that is30cases about benign tumor,31cases about breast cancer in young and32cases about breast cancer in the aged who were accepted neoadjuvant chemotherapy.2.Collect the breast cancer patients blood before neoadjuvant chemotherapy,and the1st,2nd and3rd cycles after neoadjuvant chemotherapy.Collect the blood in benign tumor before surgery;he specimen was centrifugal3000r,15min,collected supernatant,and resaved in-80℃refrigerator.3.Detected the expression about IGF-1,Caspase-3,EGFR in breast cancer patients and benign tumor patients. Measure the tumor size after neoadjuvant chemotherapy by MRI. Compare the relationship between breast cancer patients and benign tumor patients about IGF-1,Caspase-3,EGFR,and compare the relationship between before neoadjuvant chemotherapy and the lst,2nd and3rd cycles after neoadjuvant chemotherapy.Calculation the samples concentration about IGF-1,Caspase-3,EGFR in the use of regression equation,SPSS statistical packages were applied to the statistical analysis115,000.[Result] 1.Compared with benign tumor patients,IGF-1was increased in young breast cancer patients before neoadjuvant chemotherapy(P<0.05),and there was no statistics in the aged breast cancer patients before neoadjuvant chemotherapy(P>0.05); Compared with the aged breast cancer patients,IGF-1was increased in young breast cancer patients before neoadjuvant chemotherapy(P<0.05); IGF-1was decreased in young breast cancer patients after neoadjuvant chemotherapy(P<0.05);2.Compared with benign tumor patients,EGFR was increased in young breast cancer patients and in the aged breast cancer patients before neoadjuvant chemotherapy(P<0.05); EGFR was decreased in young breast cancer patients and in the aged breast cancer patients after neoadjuvant chemotherapy(P<0.05).3.Compared with benign tumor patients,Caspase-3was decreased in young breast cancer patients and in the aged breast cancer patients before neoadjuvant chemotherapy(P<0.05); There has statistics compared the young breast cancer patients and the aged breast cancer patients; EGFR was increased in young breast cancer patients after neoadjuvant chemotherapy(P<0.05); And EGFR was decreased in the aged breast cancer patients after neoadjuvant chemotherapy(P<0.05).[Conclusion]The experiment results show that:1.The expression of IGF-1was increased in breast cancer patients and decreased after neoadjuvant chemotherapy,explaining that IGF-1may be have some relationship with the occurrence and invasive in breast cancer.The young breast cancer invasive rate was higher than the aged breast cancer patients may be the case of that IGF-1was increased in young breast cancer patients,the decreased after neoadjuvant chemotherapy may be correlation with the efficacy of chemotherapy.2.The expression of EGFR was increased in breast cancer patients and it is higher in young breast cancer patients compared with the aged breast cancer patients,and it is decreased after neoadjuvant chemotherapy,explaining that EGFR may be have some relationship with the occurrence and invasive in breast cancer.The young breast cancer invasive rate was higher than the aged breast cancer patients may be the case of that EGFR was increased in young breast cancer patients,the decreased after neoadjuvant chemotherapy may be correlation with the efficacy of chemotherapy.3.The expression of Caspase-3was decreased in breast cancer patients,and it is lower in young breast cancer patients; It is increased in young breast cancer patients after neoadjuvant chemotherapy,but the highest was still lower than the aged breast cancer patients; And the expression of Caspase-3was decreased after neoadjuvant chemotherapy; Though review the literature,that different research have different reports,age may be associated with Caspase-3.So,the inhibitor of apoptosis was more significant in young breast cancer patients compared with the aged breast cancer patients.There may be correlation with the efficacy of chemotherapy.4.There was a negative correlation compared IGF-1,EGFR and Caspase-3.IGF-1,EGFR was decreased and Caspase-3was increased,the development of breast cancer may be have some relation ship with them and can be used as indicators of the evaluation of chemotherapy efficacy. |
